Abstract

目的: 构建由癌胚抗原启动子(CEApromoter,Cp)驱动的双自杀基因CD-TK治疗载体pcDNA3.1(-)Cp-CD-TK,研究癌胚抗原启动子能否控制CD-TK基因在CEA阳性大肠癌细胞中专一性表达和杀伤,比较融合基因与单基因对肿瘤细胞生长抑制方面有无差异,是否存在协同效应,并确定安全有效的药物使用浓度。观察旁观者效应。 方法: 分别以提取的人血细胞基因组DNA,pMD18-CD质粒和tgCMV/Hy-TK质粒为模板,PCR扩增Cp、CD和TK基因片段,采用双酶切、连接等方法依次将Cp、CD、TK基因亚克隆到载体pcDNA3.1(-),构建含Cp启动子和CD-TK融合基因的...Objective: To construct targeted plasmid vector pcDNA3.1(-)Cp-CD-TK which contains CEA promoter.
