Cloning and Preliminary Study of Aegiceras corniculatum AcTPC1 Gene

Abstract

Ca2+通道在高等植物细胞中参与众多的信号转导过程并起着重要的作用。随着对Ca2+通道研究的深入,越来越多不同类型的Ca2+通道陆续展示在人们的面前。2000年Ishibashi等在家鼠(rat)中发现了一种新型的Ca2+通道――TPC1(TwoPoreChannel1),经研究证明它是一种电压调控的Ca2+通道。自发现TPC1基因以来,先后在拟南芥、水稻、烟草和小麦等植物中也克隆到了TPC1基因。本文采用桐花树等材料,对这种新型的Ca2+通道基因进行克隆和研究。 从桐花树等植物中提取基因组DNA,再根据已知的几种TPC1基因保守区域设计简并引物,用PCR法成功地从桐花树等的基因组中扩增出基...Ca2+ channels participate in and play a important role in signal transduction in high plant cells.
