Isolation and Identification of a Strain of Halomonas sp.and its DAPDC Gene (lys A) Clone and Expression

Abstract

lysA基因编码二氨基庚二酸脱羧酶(DAPDC),该酶将二氨基庚二酸(DAP)转化为L-Lys,因此DAPDC对细菌的生长和生存至关重要。本实验从盐生杜氏藻高盐培养液中分离纯化了一株盐单胞菌,并从形态、生理以及16SrDNA等方面进行初步鉴定,将它命名为Halomonassp.ZL-4。 采用鸟枪法构建了Halomonassp.ZL-4部分基因组文库。提取Halomonassp.ZL-4的总DNA,经限制性核酸内切酶Sau3AI消化后回收1~6kb大小的酶切片段,与经限制性核酸内切酶BamHI酶切消化、回收后的质粒pUC19连接,热转化感受态细胞E.coliDH5α。在此基础上,进行克隆子的...DAPDC is a vitamin B6-dependent enzyme that stereospecifically converts meso-DAP to L-lysine. The enzyme is of interest because of its importance in bacterial growth and survival. Lysine is essentially required in protein biosynthesis and for their viability and development.
