Molecular analysis of bacterial diversity in the sediment of shrimps culture environment and research on some optimization methods for culturable bacteria

Abstract

传统的微生物纯培养技术能够分离得到的微生物仅为自然界中微生物总数的1%左右,绝大多数微生物还处于未培养和未研究状态,针对新型培养技术的探索已经成为了研究热点。本论文在传统微生物培养技术的基础上进行改进,对泉州湾滩涂虾池表层沉积物中的细菌进行富集培养,并运用变性梯度凝胶电泳(DGGE)技术对培养后的细菌多样性变化进行比较和研究;同时,对虾池表层沉积物样品总DNA以及多种固体培养基富集后培养物的总DNA的DGGE图谱中16SrDNA片段的显著条带进行切割、克隆和测序,并进行系统发育分析。本研究取得了以下主要结果: 1.对虾池表层沉积物细菌多样性进行分析,得出该沉积环境细菌多样性指数为3.4971...Less than 1% of the microorganisms have be cultured and isolated by traditional pure culture technique, most of them in nature remain uncultured.
