现已明确FT基因在拟南芥光周期诱导开花途径中起着关键作用。本研究以模式植物拟南芥ft-10突变体与野生型为材料,通过双向电泳技术和质谱分析等实验手段,研究了二者蛋白质水平上的差异表达,有助于深入研究FT调节光周期开花的作用机制。研究结果如下: (1)通过优化植物蛋白质提取方法及与之匹配的蛋白质裂解液,筛选出一种适合拟南芥叶片全蛋白质分析的双向电泳制样和分析系统:以醋酸铵/甲醇沉淀法提取叶片蛋白质样品,经裂解液[8mol/L尿素,2mol/L硫脲,4%CHAPS,60mmol/LDTT,2%Ampholine,1mmol/LPMSF]裂解后按1.5mg上样,经考染后获得背景清晰,蛋白分辨率较高...It is well known that FT gene plays an important role in photoperiod pathway regulating flowering in Arabidopsis.
