Synthesis of Novel Bicycloβ-Lactams As Potential β-Lactamase Inhibitors

Abstract

β-内酰胺类抗生素是临床上最常用和品种最多的一类抗生素,它们为保护人类的健康、延长寿命发挥了巨大的作用。但是,随着β-内酰胺类抗生素的广泛使用,细菌逐渐对其产生了耐药性,从而使得一些β-内酰胺药物的疗效大大下降。其中β-内酰胺酶的产生是细菌对该类抗生素产生耐药性的主要机制。研究发现,在对待由产生耐药性的细菌所引起的感染时,将β-内酰胺酶抑制剂与降低疗效的β-内酰胺类抗生素联合使用,会保持甚至加强β-内酰胺类抗生素的抗菌活性。因此,积极开发新的β-内酰胺类抗生素和高效、广谱的β-内酰胺酶抑制剂是合成化学和药物化学工作者面临的重要课题之一。本论文的目的之一是以2-金刚烷酮为原料,利用氯磺酰异腈酸酯...The β-lactam antibiotic is the most important class of antibiotic used in the clinical practice for the treatment of bacterial infections.
