Preparation of 10-hydroxycamptothecin loaded PLA microbubbles and evaluation of bioeffect on hepatocellular carcinoma cells induced by ultrasound

Abstract

摘要 癌症是威胁人类健康的重大疾病之一,恶性肿瘤的治疗在很大程度上仍以化疗为主。近年来,载药微泡联合超声辐照治疗肿瘤也日益受到重视。 羟基喜树碱(HCPT)是一种广谱的抗肿瘤药物,临床上主要剂型为注射用冻干粉。聚乳酸(PLA)有良好的生物相容性和生物降解性,在生物医学领域有广泛的应用前景。为了增加靶向部位血药浓度,降低给药量,减小毒副作用,本课题探索载羟基喜树碱-聚乳酸微泡的制备及超声辐照下对肿瘤细胞生物效应的影响。 本文的主要研究包括: (1)以聚乳酸为成膜材料,对可能影响空白微泡形貌的各个参数进行单因素考察,优化出最佳微泡设计方案; (2)以羟基喜树碱为模型药物,制备载羟基喜树碱...Abstract Cancer is one of chief diseases to threat the health of human being.
