Abstract

本论文侧重研究魟鱼肝(LiverferitinofDasyatisakajei,DALF)铁蛋白分离纯化、电泳行为、质谱特性和部分一级结构。主要研究内容如下:根据DALF抗逆性较强特点,采用热变性法、离子交换层析技术(DEAE-52)和天然聚丙烯酰胺凝胶电泳技术(PAGE)制备电泳纯铁蛋白。选用维生素C还原法,研究电泳纯DALF在30℃到45℃之间进行的释放铁全过程和动力学特性;比较在室温但不同pH(5.0-8.0)范围内,弱酸碱介质对DALF释放铁的影响。其结果有助于理解反应温度和pH对铁蛋白壳柔性调节速率的影响。在低pH介质中,采用电子显微镜技术阐明DALF亚基解离规律,了解蛋白壳分解和铁...This thesis is mainly studied on the purification、 electrophoresis behavior、 characteristic of mass spectrometry and amino acid sequence partially of the Ferritin isolated from the liver of fish named Dasyatis akajei.
