目的: 1.检测肝癌细胞耐药株模型的耐药性。 2.检测ERK1/2信号通路的特异性抑制剂索拉菲尼和PD98059对肝癌耐药细胞株耐药性的影响,以明确下调ERK1/2活性是否可以逆转肝癌细胞耐药性。 3.检测经典耐药逆转剂环孢霉素A对ERK1和ERK2表达水平的影响,初步探索ERK1/2信号通路与逆转肝癌细胞耐药性之间的关系。 方法: 1.使用阿霉素对人肝癌细胞株SMMC-7721和BEL-7402进行大剂量冲击方法诱导得到肝癌耐药株模型。采用Celltiter-Glo荧光细胞活性检测方法进行药敏实验并计算耐药指数,确定耐药细胞株的耐药性。 2.用MTT法检测索拉菲尼、PD98059...Objective 1.
