Theoretical Study on metal selectivity in Metal Regulatory Protein NikR

Abstract

镍调节蛋白(NikR)是一类响应转录因子,在细胞内镍过量下可调节生物体内镍离子转运蛋白的表达。现在实验研究表明除了和镍结合外,NikR还可以和铜、锌等其他过渡金属结合,得到相应的金属结合蛋白。这些金属离子结合NikR蛋白的亲和性大小服从过渡金属复合物稳定性Irving-Williams序列:Mn(II)Zn(II)。 为了深入理解NikR对过渡金属离子的选择性,我们除了采用简单活性中心模型,还使用量子力学/分子力学(QM/MM)组合方法和Poisson-Boltzmann方法,系统分析了蛋白环境对过渡金属结合性和选择性的影响,表明考虑蛋白环境的贡献对完全理解金属蛋白的金属结合性非常重要。计算...NikR is a nickel-responsive transcription factor that regulates the expression of nickel ion transporter in the presence of excessive concentration of intracellular nickel.
