The relationship between Kv1.3 Potassium Ion Channels and Human Osteosarcoma MG63 cells

Abstract

目的:观察和研究电压门控钾离子通道Kv1.3特异性阻滞剂rMargatoxin(MgTX)和海葵毒素(Stichodactylahelianthustoxin,SHK)对人骨肉瘤MG63细胞增殖、凋亡、细胞分裂周期、细胞迁移和侵袭能力的影响,并初步探讨其分子作用机制。 方法:体外细胞培养法培养人骨肉瘤MG63细胞株,分别用特异性Kv1.3钾离子通道阻滞剂rMargatoxin和海葵毒素(SHK)阻滞骨肉瘤MG63细胞的Kv1.3钾离子通道,采用MTT法检测不同浓度rMargatoxin和SHK对细胞增殖的影响;流式细胞术(FCM)检测其对细胞凋亡和细胞周期的影响;划痕实验检测其对MG63细胞...Objective: Many evidences have shown that voltage-gated potassium channels Kv1.3 plays an important role in the growth and development of tumor.
