Silencing of Pokemon Enhances Caspase-Dependent Apoptosis via Fas- and Mitochondria-Mediated Pathways in Hepatocellular Carcinoma Cells

Abstract

Pokemon是POK转录因子家族中的一员,具有原癌基因的活性,在多种肿瘤中均高表达,但其在肝癌细胞中的深入研究较少。我们实验室前期研究结果已经证实了Pokemon在肝细胞癌中高表达,并且通过AKT及ERK依赖性信号通路促进肝癌细胞的增殖和迁移。肿瘤的发生不仅与细胞的异常增殖和分化有关,也与细胞凋亡的异常相关。细胞凋亡有两条经典凋亡信号途径:一是死亡受体激活途径,另一条是线粒体-细胞色素C途径。两条通路的活化最终均可导致caspase通路的活化。本课题拟验证在第三代铂类化疗药物奥沙利铂的凋亡诱导下,Pokemon通过介导这两条凋亡信号途径从而参与肝细胞癌的发生发展。 本课题研究中,我们首先采...The role of Pokemon (POK erythroid myeloid ontogenic actor), a recently identified POK transcription factor with proto-oncogenic activity, highly expressed in a wide variety of tumors,but in hepatocellular carcinogenesis has only been assessed by a few studies.
