芘诱导下新鞘氨醇单胞菌差异表达蛋白追踪

Abstract
以一株芘降解新鞘氨醇单胞菌(Novosphingobium pentaromativoransUS6-1)为模式菌株,为了追踪其在有芘和没有芘诱导下差异表达蛋白,对该菌株蛋白的超声破碎提取条件进行了优化.结果表明,在4℃环境下,功率为40 W,工作时间与间隔时间均为5 s,超声总次数为60次时菌体破碎效果最佳.与IEF(Isoelectric focusing electrophoresis)自制胶相比,IPG(Immobilized pHgradient)胶分离得到的蛋白点数量更多,质量更好.分析双向电泳图谱,追踪到至少7个在芘诱导条件下菌株US6-1表达的差异蛋白点.这说明芘可以诱导菌株US6-1合成特异蛋白.本实验结果为多环芳烃(PAHs)微生物代谢途径的阐明及环境中PAHs污染修复策略的制定提供了重要依据
