Keywords  quaternary ammonium surfactant; liposome; solubilization process; dynamic light scattering; ζ-potential 细胞的各种功能, 如识别、融合、胞吞、输运、渗透等都是以膜为媒介的过程. 利用表面活性剂单分子层, 双分子层及磷脂囊泡等膜模型来研究膜结构的方法称为膜模拟方法. 特别是由两亲分子形成的囊泡. 具有与细胞膜相似的封闭双层结构, 被视为膜模拟的简单理想模型[1], 广泛应用于分子识别、纳米合成以及药物传递等研究领域. 形成囊泡的两亲分子可以是天然磷脂, 单链表面活性剂, 双链表面活性剂, 甚至正负离子表面活性剂的混合体系[2]. 由天然磷脂形成的囊泡(亦称为脂质体)以其毒性低, 制备简单, 可避免药物的降解和实现靶向给药, 已被广泛作为药物载体使用, 表面活性剂则是生物膜学及给药系统的分子研究工具. 目前, 关于表面活性剂在脂质体上的吸附、渗透、和 终破坏双层结构的研究多针对阴离子或非离子表面活性剂-囊泡体系[3～7], 阳离子表面活性剂相关的报道则较少[8]. 季铵盐二聚(Gemini)表面活性剂是一类新型表面活性剂, 其应用尚处开发阶段. 本文采用 pH 7.4, 盐浓度为0.1 mol•L－1的缓冲体系, 模仿人体细胞所处的环境. 通过研究季铵盐二聚表面活性剂(C12-8-C12•2Br)以及十二烷基三甲基溴化铵(DTAB)与脂质体的相互作  No. 6 张春艳等：季铵盐表面活性剂与卵磷脂囊泡相互作用的研究 451  用, 从阳离子表面活性剂-脂质体体系的稳定性, 电动性质等方面探讨了双子表面活性剂中连接基团对脂质体稳定性的影响. 1  实验部分 1.1  实验试剂 蛋黄卵磷脂(日本, 试用药). 季铵盐二聚表面活性剂(C12-8-C12•2Br)为本实验室合成[9], 其分子结构式如下:   Tris 缓冲液(pH 7.4): 0.005 mol•L－1 Tris, 0.1 mol•L－1 NaCl 混合液用稀盐酸溶液(0.1 mol•L－1)调至 pH 7.4. 实验中所用溶液均为缓冲液体系.  十二烷基三甲基溴化铵(DTAB): 用乙酸乙酯-乙醇混合溶液重结晶三次.  NaCl, HCl, Tris, 无水乙醇, 乙酸乙酯均为分析纯. 实验用水为三次蒸馏(电导率小于 2×10－5 S/m). 所有实验均在 25 ℃下进行． 1.2  实验方法 1.2.1  卵磷脂囊泡的制备 采用乙醇注射法[10], 用微量注射器将卵磷脂的无水乙醇溶液注射到 56 ℃的 Tris 缓冲液中, 即制得单分散的脂质体(0. 5 mmol•L－1).  将上述溶液冷至室温后, 与不同浓度的表面活性剂溶液等体积混合即得表面活性剂-脂质体体系.  1.2.2  吊片法测定表面活性剂溶液 cmc 用 Sigma701 张力仪(KSV 仪器公司, 芬兰)测定C12-8-C12•2Br 及 DTAB 表面活性剂缓冲溶液的表面张力, 从而得出二者的 cmc 分别约为 0.02, 4 mmol•L－1.  1.2.3  浊度的测定 表面活性剂-脂质体混合溶液充分振荡后 , 用Cary50 Win/UV 分光光度计(Varian 仪器公司, 美国), 测定不同时间间隔下体系的吸光值. 实验中使用 1 cm石英比色池, 入射波长 380 nm.  1.2.4  流体力学直径, Zeta 电势的测定 将表面活性剂和脂质体混合 24 h后, 将此表面活性剂-脂质体体系在 3000 r/min 转速下离心 20 min, 用Zetapals 电势仪(Brookhaven 仪器公司, 美国)在激光波长 660 nm, 固定散射角 90°下测定其流体力学直径和Zeta 电势.  2  结果与讨论 2.1  增溶作用 囊泡向胶团的转变过程中, 卵磷脂的溶解度将发生很大的改变, 因此观测溶液的浊度变化可以捕捉到卵磷脂相转变及聚集态的信息. 图 1(A)表明C12-8-C12•2Br迅速促进囊泡的溶解, 6 h 内囊泡溶解完全, 白色乳浊液迅速变成澄清的透明液, 浊度急剧下降. 该过程类似于阴离子和非离子表面活性剂与脂质体相互作用的情    况[11,12], 分为三个阶段(图 2): 阶段 I, C12-8-C12•2Br 分子从水相进入脂双层, 促进囊泡的吞包和融合, 形成卵磷脂-C12-8-C12•2Br 混合囊泡, 直到脂双层被 C12-8-C12• 2Br 所饱和. 在饱和点 B 处卵磷脂-C12-8-C12•2Br 混合胶团开始形成, 继续增加 C12-8-C12•2Br 的量, 则达到阶段II, 这时被C12-8-C12•2Br饱和的囊泡和卵磷脂-C12-8-C12• 2Br 混合胶团共存; 而阶段Ⅲ则不再有囊泡存在. 其特征是卵磷脂-C12-8-C12•2Br 混合胶团与自由的非缔合的C12-8-C12•2Br 分子达成平衡. 图 2 中 A 点处囊泡开始生长. C12-8-C12•2Br-脂质体体系以 3 mmol•L－1, 6 mmol•  L－1 为分界点划分出与图 1 相对应的“三阶段” [图3(A)]: 3 mmol•L － 1 饱和吸附点处 , 电势值 大 , 6 mmol•L－1后 ζ 电势降低到一恒定值, 囊泡完全溶解. 而DTAB-脂质体体系 ζ 电势随 DTAB 浓度的增大持续上升, 在实验浓度范围内未有转折点出现[图 3(B)]. ζ电势的结果表明表面活性剂分子进入脂双层将相应地提高囊泡的表面电势, 且 ζ 电势 大值与使双层饱和所需的有效表面活性剂浓度是相关联的[14] (表 2). 可见, 要使DTAB 在脂质体上达到饱和吸附, 其浓度应大于 10 mmol•L－1. 2.3  连接基影响 对卵磷脂囊泡 C12-8-C12•2Br 表现迅速、强烈的破坏作用, 源于联接基的存在使其头基体积较大, 进而影响其在脂质体中的吸附行为. 定义亲水头基总体积Vhead为用来构成亲水层的离子头基和被水润湿的那些亚甲基的总体积, 可得 C12-8-C12•2Br 的 Vhead 约为 DTAB 的 3倍[15] (表 3). 表 3  C12-8-C12•2Br 和 DTAB 的 Vhead 理论计算值 Table 3  Vhead of C12-8-C12•2Br and DTAB  DTAB C12-8-C12•2Br Vhead/nm3 0.168 0.476  极性物质总是将头基固定在囊泡的极性基或离子型基团层中, 它们的非极性的碳氢基则存在于囊泡的碳氢微区中[16]. 由于 Vhead大, 当单位体积内 C12-8-C12•2Br分子增多时, 空间效应使得 C12-8-C12•2Br 的极性基与卵磷脂的较大极性基不易处于同一平面内, 而采取“错位”排列方式[17](如右图所示), 使得囊泡的 外层为同电性极性基所覆盖, 囊泡之间产生一定的静电斥力; 且同电性极性基处于同一平面内, 相距很近, 也将产生一定的静电斥力. 因此造成能量的不稳定, 囊泡结构瓦解(表1). 同时, 这种排列方式使囊泡表面为 C12-8-C12•2Br 正电性头基所覆盖, 因此 ζ电势迅速上升[图 3(A)]. DTAB的 Vhead 则小得多, 其极性基与卵磷脂头基处于同一平面, 这降低了囊泡表面的负电性, 因此随 DTAB 浓度的增加, ζ 电势上升速度较 C12-8-C12•2Br-脂质体体系慢(图 3). 72 h 未见 DTAB-囊泡形态显著变化. DTAB 分子有可能“钻穿”到囊泡内部, 采取头基与脂双层的极性内表面结合的方式, 而不破坏双层结构.   囊泡到胶团的转变过程存在着许多中间形态 [18], 如蠕虫, 条状等球形囊泡的变形体, 亦或囊泡间相互交织成链珠状, 网状等. DTAB-脂质体体系溶解曲线(图1), 动态光散射的粒径变化趋势(表 1)均表明该体系仍处于过程的某一中间态, 具体形态需要电镜进一步证实.  二聚体表面活性剂通过连接基团将两个表面活性剂“单体”在离子头基处以化学键形式连接在一起[19], 其在改变膜结构方面表现出特殊的性质 . 实验中C12-8-C12•2Br 比 DTAB 更易使脂双层溶解, 少量的双子表面活性剂就能改变脂质体表面荷电性质. 因此, 可通过控制季铵盐二聚表面活性剂中连接基的长度, 来调节脂质体的药物释放速度.
