Abstract

Microsatellite instability revealed by altered pattern at STR loci is a new molecular marker that might define transformation of hemopoietic cells. A change in the number of core repeats within polymorphic sequences is caused by an increased rate of mutations of mismatch repair genes. On the other side, loss of one allele at STR locus which maps a tumor suppressor gene indicates its decreased activity or even inactivation leading to selective advantage of malignant clone. MI were longitudinally studied in different FAB subtypes of AML and MDS at various stages of the disease. The attention was paid especially to the telomere of 11 p because if contains at least three tumor suppressor regions. Genomic DNA was extracted from mononuclear and granulocyte fractions of bone marrow.
