Entwicklung einer Pruefvorschrift zur Erkennung von synergistischen Wirkungen zwischen membranschaedigenden Substanzen und anderen Schadstoffen

Abstract

A method was developed to quantify toxic combination effects of environmental chemicals. The assumption that a membrane damaging substance A enhances the cytotoxic effects of a substance B was confirmed by our experiments with human fibroblasts. Membrane damaging and cytotoxic effects correlated well with their log P_O_W-values (up to r=0.99, p<0.01). The cytotoxic effects correlated well with the membrane damage (up to r=0.99, p<0.01). Quantification of synergistic effects was obtained by comparing growth inhibition induced either by substance B alone or in combination with subtoxic concentrations of substance A. The log of the 'synergistic combination factor' ([EC_2_0_s_u_b_s_._B-EC_2_0_s_u_b_s_._A_+_B]/c_s_u_b_s_._A) correlated well with the log P_O_W-value of substance A (r=0.97, p<0.01). The subtoxic concentration of substance A needed for enhancement of growth inhibition induced by substance B by a factor 2 was obtained using the equation log c=-0.56.log P_O_W_._s_u_b_._A+1.90.
