Mise au point du dosage de 8 organophosphores en GC/MS (application à un cas d'intoxication volontaire au dichlorvos)

Abstract

Nous avons mis au point et validé une technique de quantification simple, rapide et reproductible de 8 organophosphorés en GC/MS. Les échantillons sont traités par extraction liquide/liquide. Le rendement d'extraction des différentes molécules varie de 20 à 123%. Cette méthode présente une exactitude comprise entre 97 et 117,4%. Les coefficients de variation intra et inter journaliers sont inférieurs à 15%. Nous avons appliqué la technique décrite précédemment aux différents prélèvements ont nous disposions et provenant d'une personne décédée suite à une ingestion volontaire de dichlorvos. Des concentrations très importantes de DDVP dans le sang cardiaque (4,35æg/ml) t dans le cœur (1,4mg/ml) ont été observées. Les concentrations du sang périphérique et des trines sont de 1,3 æg/ml et celle du foie est indétectable. La quantité retrouvée dans le foie est le 140g. Nous rapportons le premier cas français d'intoxication et de distribution tissulaire de dichlorvos.A simple, rapid and reproductive method for the quantification of 8 OP pesticides in human biological matrice, using GC/MS is presented. The biological assay were extracted using a liquid/liquid extraction. Extraction recovery were in the ranges between 20 and 123%. The accuracy of the procedure were ranged from 97 and 117,4%. This method is found repeatable and yield good intra and inter precision (respectively .< 14,8% and < 15%) for all compounds. .. The different blood levels and tissues distribution of DDVP are determined in a case of fatal self poisoning using the method described above. High concentration .are found in cardiac blood and heart (respectively 4.35 æg/ml and 1.4 mg/ml). The concentration of DDVP in peripheric blood and urine are 1.3 æg/ml and the quantity of the liver is not detectable. The remaining DDVP amount in the stomach was calculated as 140 g.
