ADE occurs when non-neutra-lised virus-antibody complexes find alternative receptorsand routes entry into the cell via the Fc-receptor path-way. ADE has been demonstrated predominantly inmacrophages or Fc-receptor bearing cells although othertypes of cells have also been occasionally implicated(1,2). Thus, viruses may find routes of entry to cellslacking the usual virus receptor. Alternatively, the innateimmune signals triggered by virus infection via the FcRpathway may be different to those triggered by entry viathe physiological virus receptor. Both these conse-quences may have implications for virus tropism andpathogenesis. Recently there have been reports ofincreased infection rates by pandemic influenza H1N1virus following receipt of seasonal flu vaccination (3,4).ADE has been proposed as one mechanism. Here weillustrate the possible role played by humoral immunityin providing Influenza A viruses an opportunity to bet-ter infect immune cells.Our results showed that, in some cases, prior additionof human serum to the inoculum triggered an enhancedinfection of target cells as illustrated by a ~2-5 foldincrease in Influenza M-gene copy numbers. Immuno-fluorescent microscopy revealed that serum-mediatedpdmH1N1 infection led to a higher number of infectedcells. As the fold increase of infected cells paralleled thefold change in viral gene copies, we conclude that ADEwas acting by increasing the number of infected cellsrather than solely increasing the viral load per cell.Based on their ability to enhance pdmH1N1 infectiv-ity, human sera could be divided into 3 distinct groups:some showing neutralization of infection, others increas-ing the yield of pdmH1N1-infected cells. The thirdgroup were those with no or marginal increase of virusinfection, one which was not sustained over the testedrange of serum dilutions.Our results demonstrate that the newly emerged pan-demic H1N1 Influenza A virus infection of cells of thehematopoietic lineage may be enhanced by the presenceof some human sera.
