Amino acid analysis of P. m onodon  muscle

Knowledge o f the amino acid pattern o f P. monodon is necessary in ascertaining whether 
the feed meets the optim um  amino acid pattern requirements fo r growth. This study was 
undertaken to  gain benchmark inform ation on the protein quality o f the feeds to  be used.
Two adult female prawns weighing 33.84 g (Prawn I ) and 38.71 g (Prawn II) were used fo r 
the analysis. The muscle tissue was chopped, dried at 110°C to  constant weight and ground. 
A  20 mg sample was taken fo r protein analysis by the micro-Kjeldahl method and another 20 mg 
was hydrolyzed under vacuum in 6H HCI fo r 24 and 48 hr fo r amino acid assay. A  separate 20 mg 
sample was hydrolyzed in 6N NaOH fo r  24 hr fo r tryptophan analysis. The Yanaco Model LC-5S 
amino acid analyzer was used fo r the analysis.
Values obtained fo r white tuna meat are also shown fo r comparison (Table 1). The results 
show that amino acid values o f Prawn I and Prawn II are fa irly  close, w ith  some differences 
obtained fo r proline, alanine and lysine. However, on the whole, the analysis shows similar 
patterns. Comparing w ith  tuna meat which is used as feed fo r P. monodon postlarvae, significantly 
lower values were obtained fo r arginine, proline, lysine and aspartic acid. Cystine has low 
concentration in tuna meat and is not detected in prawn. Ammonia is derived from  glutamine 
and asparagine. Thus, the tota l concentration o f glutamic acid and aspartic acid, in micrograms 
per 100 micrograms of prote in, is 11.12 fo r tuna, 18.63 fo r Prawn I and 18.58 fo r Prawn II.
