Dissertation Nora Grünewald                          Abstract  Abstract Higher brain function requires precise processing of excitatory and inhibitory signals. In spinal cord and brain stem the inhibitory signal transmission is primarily based on the activation of glycine receptors (GlyRs). The exclusive interaction of the GlyR β-subunit with the major scaffolding protein of inhibitory synapses, called gephyrin, highlights the crucial role of this subunit for the postsynaptic confinement of hetero-pentameric GlyRs. Synaptic plasticity events at glycinergic synapses are predominantly initiated by modulating GlyR synaptic diffusion to regulate receptor numbers at the synapse and can be exerted by affecting receptor/scaffold interactions. The underlying molecular mechanisms still remain elusive, especially regarding GlyR binding by gephyrin in a pentameric assembly of the receptor. The first part of this study characterized the thermodynamic parameters of GlyR β-loop (βL) and gephyrin interaction to understand the bimodal binding of both proteins. Following the truncation of the βL from the N- and C-terminal ends it was possible to dissect the low- and high-affinity binding of βL to gephyrin and elucidate the requirements for a biphasic interaction. Additional regions of interaction on gephyrin were identified via chemical crosslinking and found to undergo contacts apart from the canonical binding pocket. High-affinity binding was found to depend on the C-terminal folding of the βL and was identified to be critically important for the synaptic confinement of GlyRs by gephyrin in spinal cord neurons. Stabilization of synaptic GlyRs by postsynaptic gephyrin clusters is very dynamic and can directly affect the amount of receptors at the synapse. The interaction of both proteins is modulated on short-time scale by phosphorylation, which represents an important mechanism of synaptic plasticity. A dispersal of synaptic GlyRs upon protein kinase C (PKC) activation was identified and due to the phosphorylation of βS403 thus resulting in a significant reduction in gephyrin binding affinity. Based on isothermal titration calorimetry (ITC) studies, it was possible to unravel that one binding site disappears following βS403 phosphorylation and causes a reduced affinity of the remaining one. In contrast, phosphorylation of βY413 was found to potentially participate in the acceleration of GlyR function upon protein tyrosine kinase (PTK) activation by increasing the specific binding to gephyrin, probably resulting in a decreased receptor off-rate.  So far, studies on receptor and gephyrin interaction were conducted with isolated GlyR peptides. Therefore, the interaction of gephyrin with GlyR β-loop as well as various subtypes of the GABAAR should be improved by establishing a model system based on yeast pentameric lumazine synthase (LS). Homology studies using the nicotinic acetylcholine receptor (nAchR) suggested that the large cytoplasmic domains of a GlyR, when inserted into LS, would be able to resemble the physiological arrangement within holo-receptors. The here generated LS-GlyR-loop chimeras interacted via multiple β-loops with gephyrin based on high affinity and were able to link several gephyrin molecules to build high–molecular weight complexes. It was hypothesized that receptor and gephyrin contain the intrinsic property of complex formation, which provides an entirely novel concept on the postsynaptic assembly and stabilization at glycinergic synapses.  Dissertation Nora Grünewald                  Zusammenfassung  Zusammenfassung Die sensorische und motorische Reizverarbeitung im Rückenmark und Hirnstamm erfordert die genaue Abstimmung der erregenden und hemmenden Signale und basiert hauptsächlich auf der Aktivität von Glycin Rezeptoren (GlyRen). Deren synaptische Lokalisation wird durch die exklusive Interaktion der GlyR β-Untereinheit mit Gephyrin, dem wichtigsten Netzwerk-bildenden Protein der hemmenden Synapse, vermittelt. Die Gephyrin-abhängige dynamische Diffusion von GlyR ist ein Schlüsselelement der synaptischen Plastizität, da diese einen direkten Einfluss auf die Anzahl an synaptischen Rezeptoren hat. Die zugrundeliegenden molekularen Mechanismen der Rezeptor/Netzwerk Interaktion, besonders in Bezug auf den pentameren Aufbau des GlyRs, sind noch nicht im Detail bekannt.   Der erste Teil dieser Studie befasste sich mit der Charakterisierung der bimodalen Interaktion des GlyR β-Loops (βLs) mit Gephyrin auf thermodynamischer Ebene (ITC). Durch die N- und C-terminale Verkürzungen des βLs was es möglich die hoch- und niedrig-affine Bindestelle zu identifizieren und die molekularen Voraussetzungen der biphasischen Interaktion zu beschreiben. Dabei konnte anhand von chemischer Vernetzung die Beteiligung weiterer Regionen von Gephyrin an der Binding des βLs nachgewiesen werden. Die hoch-affine Bindestelle basiert auf einer bestimmten helikalen Faltung der C-terminalen Reste des βLs und ist unabdingbar für die synaptische Stabilisation von GlyRen durch Gephyrin in Rückenmarksneuronen.  Diese Stabilisierung ist sehr dynamisch und hat einen direkten Einfluss auf die Anzahl an synaptischen GlyRen. Die Interaktion beider Proteine kann mittels Phosphorylierung beeinflusst werden ist ein wichtiger Bestandteil der Plastizität an glyzinergen Synapsen. In diesem Sinne führte eine erhöhte Aktivität von Protein Kinase C (PKC) zu einer reduzierten Anzahl an synaptischen GlyRen. Dies resultierte aus der spezifischen Phosphorylierung des βS403, was eine signifikante Reduktion der Gephyrin Bindestärke zur Folge hatte. Durch Isothermale Titrations Kalorimetrie (ITC) Studien konnte gezeigt werden, dass dieser Effekt auf den Verlust einer Bindestelle zurückzuführen ist, wobei die verbliebene Interaktion nur noch mit einer reduzierten Affinität stattfinden konnte. Im Gegensatz dazu basiert die Verstärkung der GlyR Funktion auf der Phosphorylierung von βY413 durch Protein Tyrosin Kinase (PTK) und beinhaltet auch eine erhöhte Anzahl an spezifischen Interaktionen des GlyR β-Loops mit Gephyrin, die vermutlich die Dissoziationsrate des Rezeptors stark reduzieren. Bislang wurden Interaktionsstudien von Rezeptoren mit Gephyrin ausschließlich mit isolierten Peptiden durchgeführt. Darum sollte die Interaktionsanalyse von Gephyrin mit dem GlyR β-loop oder verschiedenen Untereinheiten des GABAARs durch die Einführung eines Modellsystems verbessert werden. Dieses beruht auf der pentameren Lumazine Synthase (LS) aus Hefe. Durch Homologie-studien wurde angenommen, dass die Rezeptorloops eine nahezu physiologische Struktur annehmen können. Die hoch-affine Interaktion mehrerer β-Loops der LS-GlyR-loop Chimäre mit Gephyrin begünstigte die Bildung von hoch-molekularen Strukturen. Daraus entstand die Hypothese, dass die Komplexbildung bereits eine intrinsische Eigenschaft von GlyRen und Gephyrin ist und somit ein neuartiges Konzept der postsynaptischen Assemblierung und Stabilisierung an glyzinergen Synapsen darstellt.
