Abstract Prohibitins comprise a highly conserved and ubiquitously expressed protein family and various studies unravelled essential cellular functions in vitro and in vivo. Two family members, PHB1 and PHB2, assemble into large ring-shaped protein complexes that might form protein and lipid scaffolds in the mitochondrial inner membrane, however, their precise molecular functions are still largely unknown. Studies in mouse embryonic fibroblasts (MEFs) revealed that genetic deletion of prohibitins leads to mitochondrial fragmentation, aberrant mitochondrial ultrastructure and increased apoptotic sensitivity. Mitochondrial fusion and structure depend on OPA1, a dynamin-like GTPase located in the mitochondrial inner membrane, whose activity is proteolytically regulated. Constitutive OPA1 cleavage results in the accumulation of non-cleaved, long (L-OPA1) and cleaved, short (S-OPA1) OPA1 forms which are both required for OPA1 function. However, in response to mitochondrial stress, the protease OMA1 is activated leading to the complete conversion of long to short forms, which inhibits fusion, perturbs cristae morphogenesis and renders cells susceptible to apoptosis. In prohibitin-depleted cells, L-OPA1 forms are lost while S-OPA1 forms accumulate. Re-expression of a non-cleavable L-OPA1 form restores mitochondrial morphology and cristae structure identifying accelerated OPA1 processing, probably by OMA1, as the primary cause for the observed defects in the absence of prohibitins. Consistently, neuron-specific prohibitin-deficient (Phb2NKO) mice display selective loss of long OPA1 forms and defective mitochondrial ultrastructure associated with massive neurodegeneration which leads to premature death of the animals. To investigate whether L-OPA1 stabilisation bypasses the defects observed upon loss of prohibitins in vivo, Oma1 was genetically deleted in Phb2NKO mice. Loss of Oma1 stabilises L-OPA1 demonstrating that OMA1 is activated in the absence of prohibitins. Moreover, Oma1 ablation prolongs the lifespan of Phb2NKO mice by extending neuronal survival and stabilises mtDNA levels. However, despite sustained L-OPA1 levels, Oma1 deletion in Phb2NKO mice does neither restore cristae structure nor the assembly of respiratory chain supercomplexes. Thus, these defects are rather direct consequences of the lack of prohibitins than caused by loss of L-OPA1. Functional analyses in prohibitin-depleted cells confirmed that deletion of Oma1 stabilises L-OPA1 and revealed that this confers resistance to H2O2-induced apoptosis. Nevertheless, in line with the observations made in Oma1-deficient Phb2NKO mice, cells lacking both prohibitins and OMA1 display mitochondrial fragmentation and an altered mitochondrial cristae structure along with severely impaired assembly of respiratory chain supercomplexes. Thus, this work provides novel insight into prohibitin functions by dissecting L-OPA1-dependent as well as L-OPA1-independent cellular consequences observed upon loss of prohibitins in vivo and in vitro.   Zusammenfassung Prohibitine stellen eine hochkonservierte und ubiquitär exprimierte Familie von Proteinen dar, die bereits mit einer Vielzahl von essentiellen zellulären Prozessen in Verbindung gebracht wurde. Zwei Vertreter, PHB1 und PHB2, bilden große ringförmige Komplexe in der inneren mitochondrialen Membran. Es wurde zwar postuliert, dass diese als Protein- und Lipidgerüst dienen, ihre molekularen Funktionen sind jedoch größtenteils unbekannt. Untersuchungen in murinen embryonalen Fibroblasten (MEFs) ergaben, dass die genetische Deletion von Prohibitinen zur Fragmentierung des mitochondrialen Netzwerks, einer veränderten mitochondrialen Ultrastruktur und erhöhter apoptotischer Sensitivität führt. Mitochondriale Struktur und Dynamik hängen von OPA1, einer dynamin-verwandten GTPase in der Innenmembran, ab, deren Aktivität proteolytisch reguliert wird. Unter normalen Umständen führt die Prozessierung von OPA1 zur Ansammlung von langen (L-OPA1) und kurzen (S-OPA1) OPA1 Formen. Unter Stress wird jedoch die OMA1 Protease aktiviert, was den Verlust langer und die Anreicherung kurzer OPA1 Formen zur Folge hat. Dies verhindert wiederum mitochondriale Fusion, führt zu abnormaler mitochondrialer Ultrastruktur und erhöht die apoptotische Sensitivität. Interessanterweise weisen Prohibitin-defiziente Zellen eine erhöhte OPA1 Prozessierung, vermutlich durch OMA1, auf und die gezielte Expression einer langen OPA1 Form kann sowohl die mitochondriale Morphologie als auch die Ultrastruktur wiederherstellen. Neuronen-spezifische Prohibitin-defiziente (Phb2NKO) Mäuse zeigen ebenfalls eine erhöhte OPA1 Prozessierung und veränderte mitochondriale Ultrastruktur. Massive Neurodegeneration führt zum frühzeitigen Tod der Tiere. Um zu untersuchen, ob, ähnlich wie im zellulären System, die Stabilisierung von L-OPA1 zu einer Verbesserung der beobachteten Phänotypen führt, wurde Oma1 in Phb2NKO Mäusen genetisch deletiert. Zunächst wurde beobachtet, dass der Verlust von OMA1 tatsächlich zur Stabilisierung von L-OPA1 führt. Bemerkenswerterweise verlängerte Oma1 Depletion die Lebensdauer von Phb2NKO Mäusen und stabilisierte mitochondriale DNA Level. Jedoch konnte weder eine Verbesserung der mitochondrialen Struktur noch der Assemblierung von respiratorischen Superkomplexen festgestellt werden. Dies lässt darauf schließen, dass diese Defekte durch den Verlust von Prohibitinen und nicht durch den Verlust von L-OPA1 hervorgerufen werden. Funktionelle Analysen in Prohibitin-defizienten MEFs bestätigten die Stabilisierung von L-OPA1 durch die Depletion von Oma1 und zeigten, dass dies vor H2O2-induzierter Apoptose schützt. Jedoch, im Einklang mit Beobachtungen in Oma1-defizienten Phb2NKO Mäusen, wiesen diese Zellen weiterhin fragmentierte Mitochondrien, eine veränderte Cristaestruktur und eine beeinträchtigte Assemblierung von Atmungskettenkomplexen auf. Auf Grundlage dieser Befunde lassen sich somit L-OPA1 abhängige sowie unabhängige Funktionen von Prohibitinen unterscheiden.
