Determination of specific migration of the plastici zers di-(2-ethylhexyl) phthalate and di-(2- ethylhexyl) adipate from flexible PVC films to fatt y foods: Method validation and sanitary control of flexible PVC film

Abstract

O poli (cloreto de vinila) - PVC tem sido amplamente utilizado na indústria de embalagens, na forma de filme flexível de PVC, parao acondicionamento de alimentos. Porém, para que seja possível esta utilização é necessária a adição de plastificantes. Dentre os plastificantes mais utilizados em filmes flexíveis de PVC estão o ftalato de di-(2-etil-hexila) – DEHP e o adipato de di-(2-etil-hexila) - DEHA. O DEHP é classificado pela Agência Internacional para Pesquisa em Câncer - IARC no grupo 2B (possível agente carcinogênico ao homem), e pelaAgência de Proteção Ambiental dos Estados Unidos da América - U.S.EPA no grupo toxicológico B2 (provável carcinógeno humano), sendo relacionado a diversos efeitos tóxicos para a saúde humana, tais como danos causados nos sistemas reprodutivos masculino e feminino e efeitos na indução de hepatocarcinomas. O DEHA é classificado pela IARC no grupo 3 (não classificável quanto à sua carcinogenicidadepara os seres humanos). Contudo, a U.S.EPA classifica o DEHA dentro do grupo toxicológico C, como um possível carcinógeno humano. Os dois plastificantes estão relacionados à indução da proliferação de peroxissomas hepáticos e danos à gestação. A Resolução nº 17 da ANVISA de 17 de março de 2008 estabelece limite de migração específica para DEHP em 1,5 mg kg-1do solvente simulante e para o DEHA em 18 mg kg-1. A Resolução nº 51 da ANVISA de 26 de novembro de 2010 estabelece condições para o ensaio de migração, porém é necessária a validação intralaboratorial do método. Com o objetivo de preencher esta lacuna, o presente trabalho apresenta o desenvolvimento e a validação intralaboratorial de um método para avaliação da migração dos plastificantes DEHP e DEHA da embalagem para alimentos gordurosos. O teste de migração foi realizado por meio do contato de 1 dm2do filme de PVC com 100 mL de solvente simulante etanol 95 % por 48 h à 20 ºC. Asconcentrações de migração dos plastificantes DEHP e DEHA foram determinadas por cromatografia a gás com detecção por ionização em chama e coluna de sílica fundida recoberta internamente com fase estacionária constituída de 5% fenilmetilsilicone. O método foi validado intralaboratorialmente e considerado adequado ao propósito. As faixas de trabalho foram definidas de 0,6 a 10,0 mg kg-1para o DEHP e 2,5 a 40,0 mg kg-1 para o DEHA. As curvas se apresentaram estatisticamente lineares, não sendo observada falta de ajuste ao modelo. Não foi observado efeito de matriz e a seletividade foi considerada satisfatória. O método apresentou repetibilidade e precisão intermediária adequadas e resultados robustos para temperatura e tempo de migração de (20 ± 2) ºC e (48 ± 0,5) h, respectivamente. Trinta e sete amostras de filme de PVC foram ensaiadas, apresentando resultados para a migração específica de DEHA entre não detectável (<0,35 mg kg-1) e 231 mg kg-1de solvente simulante e entre não detectável (<2,23 mg kg-1) e 304 mg kg-1de solvente simulante para o DEHP. Dentre as amostras ensaiadas, 95 % apresentaram resultado insatisfatório para pelo menos um dos plastificantes.PVC has been widely used in the packaging industry,in the shape of flexible PVC film, for packaging foods. However, for this use is required the addition of plasticizers. Among the plasticizers commonly used in flexible PVC films are di-(2-ethylhexyl) phthalate – DEHP and di-(2-ethylhexyl) adipate – DEHA. DEHP is classified by the International Agency for Research on Cancer - IARC in Group 2B (possibly carcinogenic agent to humans) and by US Environmental Protection Agency – U.S.EPA in group B2 (probable carcinogenic agent tohumans), being related to many toxic effects to human health, such as damage to the male and female reproductive system and effects on hepatocellular carcinoma development. DEHA is classified by IARC in Group 3 (not classifiable as to its carcinogenicity to humans). However, the US Environmental Protection Agency – U.S.EPA classifies DEHA within the toxicology group C as a possible human carcinogen. Both plasticizers are related to the induction of hepatic peroxisome proliferation, and damage to pregnancy. Resolution Nº 17 of ANVISA of 17 March 2008 establishing specific migration limit for DEHP in 1,5 mg kg-1of simulant solvent and 18 mg kg-1for DEHA. Resolution Nº 51 of ANVISA of 26 November 2010 establishing conditions for the migration test, but is required the in-house validation of the method. Aiming to fill thisgap, this paper presents the development and in-house validation of a method forthe determination of the migration of DEHP and DEHA from packaging for fatty foods. The migration test was carried through contact of 1 dm2of PVC film with 100 mL of simulant solvent ethanol 95 % for 48 h at 20 °C. The migration concentrations of DEHPand DEHA were determined by gas chromatography with flame ionization detector and a fused silica column coated internally with a stationary phase consisting of 5%phenylmethylsilicone. The method was in-house validated and considered suitable for purpose. The working ranges were defined from 0.6 to 10.0 mg kg-1for DEHP and from 2.5 to 40.0 mg kg-1for DEHA. The curves were statistically linear, with no observed lack-of-fit. There was no matrix effect and the selectivity of the method was satisfactory.The method has adequate repeatability and intermediate precision and robustresults for temperature and time migration of (20 ± 2) °C and (48 ± 0,5) h, respectively. Thirty seven PVC film samples were tested, presenting results for specific migration of DEHA between not detectable (<0.35 mg kg-1) and 231 mg kg-1of simulant solvent and between not detectable (<2.23 mg kg-1) e 304 mg kg-1 simulant solvent for DEHP.
