Abstract
利用粪便DNA技术对候鸟进行物种鉴定,对了解鸟类在繁殖地和越冬地的种类分布、研究其迁徙路线等具有重要意义。本研究扩增、测定了76份雁形目鸟类粪便的D-loop区序列,将其与NCBI数据库中已知鸟类同源序列进行比对分析(BLAST比对法)。结果表明:相似度为98.07% —100%,其中75%的单倍型可以被认定为单一物种,25%的单倍型检测为非单一物种,即包含2个近缘物种。比较基于Ts、Ts +Tv碱基替换计算的K2P遗传距离,说明后者更适合于D-loop区序列特征。而且,以此为基础的遗传距离法、ABGD划分法和系统发育树法可以得出一致的物种鉴定结果,消除了BLAST比对法时出现的非单一物种。由此构建了一套方法齐全、标准明确的鸟类粪便物种鉴定体系。该体系使得鸟类物种鉴定流程更具系统性、规范性,得出可信的鉴定结论,可以推广应用于其他检材的物种鉴定领域
