The low bacterial dose required toinfect and its potentially fatal consequences makethis organism a serious threat to the public healthin both the developed and developingcountries[2]. In poultry, the infection is oftenasymptomatic and chickens may hence carry thedisease to the age of slaughtering, effectivelycontaminating the slaughter house[2].To date, most droplet microfluidic chips arefabricated in poly(dimethylsiloxane) (PDMS)bonded to glass slides due to the ease offabrication and surface modification[3]. Thisfabrication method is, however, not applicable tomass production and the potential forcommercialisation is therefore reduced.Alternatively, injection moulding and hotembossing of thermoplastics are of interest.MethodsWater-in-oil droplets are produced by a flowfocusing chip. A PCR sample is injected into theaqueous line using a HPLC injection loop.The droplets are collected in an injectionmoulded disposable all-polymer chip which hasbeen bonded (sealed) using ultra sonic welding.This results in a turnaround time ~1 minute/chip.The chips droplet incubation chamber isdesigned to exploit the positive buoyancy of thedroplets to facilitate optimal droplet packingregardless of the droplet production rate andwater-to-oil flow-rate ratio. The height of thechamber restricts the droplets to pack into asingle layer. This monolayer enablesfluorescence read out by fluorescencemicroscopy.Droplet size and fluorescence intensity isextracted from the images using a customMATLAB script which performs a circular houghtransformation to identify droplets.Thermocycling is conducted on a flat bed PCRmachine controlled by a custom made LabVIEWscript.ResultsA single image contains as many as 20.000droplets which may be analysed all at once. Thedroplets are highly uniform and forms almostexclusively a single densely packed monolayer.Amplification by on-chip droplet PCR wasconfirmed by gel electrophoresis.ConclusionA disposable all-polymer multipurpose dropletinterogation chip has been fabricated anddemonstrated. Using a standard fluorescencemicroscope, thousands of droplets could beanalysed using a single snap-shot.
