人源饲养层对人胚胎干细胞生长的影响

Abstract
【目的】 选择最佳人源饲养层,并检测不同组织来源的人源饲养层细胞碱性成纤维细胞生长因子(bFGF)的分泌情况,探讨其水平与人胚胎干细胞(hESCs)生长是否相关65377; 【方法】 原代培养包皮真皮65380;子宫内膜基质65380;绒毛65380;输卵管65380;胎儿真皮65380;胎儿肌肉及胎鼠成纤维细胞(MEFs);按组织来源分为7组(MEFs为对照),同期接种同一株hESCs,从饲养层细胞密度65380;丝裂霉素作用时间等,寻找不同饲养层适合hESCs生长的最佳条件;按人体组织分6组,ELISA方法检测各种人源饲养层细胞分泌的bFGF65377; 【结果】 根据饲养层细胞生长特性,饲养层从优到劣依次为:包皮65380;子宫内膜65380;绒毛65380;输卵管65380;胎皮65380;胎肌;根据hESCs生长状况,饲养层排序为:包皮65380;输卵管65380;子宫内膜65380;绒毛65380;胎肌65380;胎皮65377;输卵管65380;包皮65380;绒毛65380;胎肌65380;子宫内膜65380;胎皮分泌的bFGF(pg8226;10-58226;mL-1)分别为13.23 ± 3.39,1.99 ± 0.17,1.40 ± 0.17,2.02 ± 1.59,0.38 ± 0.28,0.29 ± 0.2965377;输卵管与其它组织细胞分泌的bFGF差异均有统计学意义(P 0.05),与子宫内膜65380;胎皮差异有统计学意义(P 0.05)65377; 【结论】 包皮来源的饲养层最佳,输卵管细胞分泌的bFGF量最高,饲养层细胞自身分泌的bFGF和hESCs生长无必然相关性65377
