全反式维甲酸对大鼠骨髓间质细胞的诱导分化作用

Abstract
【目的】探讨全反式维甲酸对大鼠骨髓间质细胞(BMSCs)的诱导分化作用。【方法】从大鼠骨髓中分离培养间质 细胞并传至第6 代, 诱导前24 h 加1 μg·L-1碱性成纤维生长因子(bFGF)入培养液中以促分裂, 再以反式维甲酸作诱导剂, 然 后观察细胞形态的变化, 并采用免疫组织化学法, 对诱导后第4 天的细胞行巢蛋白(nestin)、神经丝蛋白(NF)、神经元特异性 烯醇化酶(NSE)表达的检测。对第8 天的细胞行脂肪细胞特异性苏丹Ⅱ 染色。【结果】诱导后第4 天的部分细胞不仅在形态 上表现为神经元样, 而且呈nestin 、NF 及NSE 阳性;诱导后第8 天的部分细胞呈苏丹Ⅱ 染色阳性。【结论】骨髓间质细胞具有 多向分化潜能, 在反式维甲酸作用下, 可被诱导成神经元样细胞及脂肪细胞
