ENH 蛋白与病理性疼痛相关性研究

Abstract
摘 要: 【目的】 观察ENH(Enigma Homolog)蛋白在大鼠坐骨神经选择性损伤(SNI)疼痛模型引起的病理性疼痛中的表达,并探索其与疼痛的相关性及可能机制。【方法】取15只雄性SD大鼠,分为3组(每组5 只),分为SNI组,麻醉治疗组和假手术对照组。另取30只雄性SD大鼠,分为6组(每组5 只),按时间点分为非手术组对照组和实施SNI手术的1、4、7、10、14d组。通过检测大鼠50%机械刺激撤足阈值,观察SNI,SNI+利多卡因和罗哌卡因复合制剂及假手术组在4、7、10 d大鼠痛敏的改变以及SNI手术组大鼠在1、4、7、10、14d的痛敏变化。免疫荧光组织化学染色方法观察各组术后10 d相应脊髓节段内ENH蛋白的表达。蛋白质印迹法(Western Blot) 检测相应各实验组及时间点脊髓节段裂解液内ENH蛋白的表达量。【结果】与假手术组相比,SNI术后10 d大鼠手术侧痛敏明显增高(n = 5/组,P < 0.01),而SNI + 麻醉药组大鼠痛敏则与假手术组接近。免疫荧光染色与蛋白质印迹法显示SNI术后10d,脊髓L4-6节段手术侧背角内ENH表达明显高于假手术组(n = 5/组,P < 0.01);SNI +麻醉药组的ENH蛋白表达则与假手术组接近。ENH与正常对照组相比,SNI组术后手术侧痛敏、ENH蛋白及mRNA有一过性增高继而恢复正常,并随时间推移而逐渐再次增高(n= 5/组)。【结论】 ENH蛋白在大鼠慢性病理性疼痛模型中有表达,且其改变与痛敏的进程一致。这些结果为ENH可能作为一种新的疼痛分子标志物奠定了理论基础
