Abstract
【目的】 检测选择性切断腰5脊神经前根(L5-VRT)术后脊髓背角内IL-6的异常表达并评价其对运动神经损伤所致机械性痛觉过敏的影响。【方法】 在不损伤感觉传入的情况下,L5-VRT制备大鼠机械性痛过敏模型,采用Western blot和免疫荧光组织化学方法检测IL-6在脊髓背角的表达水平并进行细胞定位,通过痛行为学测试观察蛛网膜下腔内注射IL-6中和抗体对L5-VRT 大鼠机械性痛过敏的影响。分组:sham组、L5-VRT组,n = 5;vehicle+ L5-VRT、IL-6 neutralization +L5-VRT组, n = 6。【结果】 L5-VRT术后7 d较假手术组同侧DRG IL-6蛋白表达显著上调(P < 0.001),IL-6蛋白表达的细胞类型为神经元细胞,L5-VRT 术前应用IL-6中和抗体可抑制大鼠L5-VRT术后所致双侧机械性痛过敏的形成(P < 0.05)。 【结论】 IL-6参与介导L5-VRT 所致的大鼠后肢机械性痛过敏
