A1.8 kb EcoRI fragment of the plasmid was used as a probe, whichcovered the 5' portion of the gene.Polymorphisms: TaqI detected two 2-allele polymorphisms (setAa and Ab) with fragment size of 3.4 kb (Al) or 3.2 kb (A2),and 2.2 kb (Al) or 1.5 kb (A2). Constant bands of 6.6, 3.7,1.7, 1.2 and 0.8 kb were also detected.Frequencies: Studied in 65 unrelated individuals of the normalJapanese (2). The polymorphism was also observedin the Wilms' patients with the allelic frequencies being similar.Therefore, a particular allele seemed not to associate with genesisof the tumor. Twoadditional faint constant bands of 1.90 kb and 0.5 kb are alsodetected by this probe.Frequency: Allele frequencies were determined from 94 unrelatedindividuals, including 79 CEPH parent DNAs. Al = 0.26 A2= 0.74Not Polymorphic For: BanH and HindIll (2 4 individualsscreened).Chromosomal Location: Initial localization to chromosome 6 wasdetermined using somatic cell hybrids, followed by more preciseassignment to 6p23 -24 using in situ and multipoint mappingtechniques (3).Mendelian Inheritance: Co-dominant segregation was observedin 21 informative CEPH families (248 individuals) (3).Probe Availability: Request for probe to Dr.A.S.Baldwin at theabove address.Other Comments: This RFLP was observed under normalhybridization and washing (i.e. 0.1 x SSC 0.1 % SDS at 65°Cfor 15 minutes) conditions.
