Synopsis: The clinical development of anti-CD73 antibodies has demonstrated their utility, but 59 the mechanism for this effect has not been fully elucidated. We now show that targeting the 60 adenosine pathway reduces extracellular adenosine in myeloid rich, hypoxic areas of pancreatic 61 ductal adenocarcinoma, abrogating the pro-tumorigenic, immunosuppressive 62 microenvironment and improving the efficacy of anti-tumour interventions
