MARIANA LOPES DOS SANTOS; DÉVORA NATALIA RANDON; FERNANDA HENDGES DE BITENCOURT; FERNANDA SPERB LUDWIG; LUIZ FELIPE PORTELA; ANA PAULA PIZZIO BECKER; GUILHERME MENEGUZZI PAGEL; BRUNA BENTO; FERNANDA SALES LUIZ VIANNA; IDA VANESSA DOEDERLEIN SCHWARTZ HCPA - Hospital de Clínicas de Porto Alegre  Introduction: Medium-chain acyl-CoA dehydrogenase deficiency (MCADD) is a fatty acid oxidation disorder (FAOD) caused by pathogenic variants in the ACADM gene, being c.985A>G (p.Lys329Glu) the most common one, accounting for approximately 90% of the mutant alleles in European patients clinically identified. Since FAOD are not included in the Brazilian neonatal screening program, population-based studies of prevalence are essential to early diagnosis, treatment and neonatal screening considerations. Objectives: To assess the frequency of c.985A>G (ACADM) in a healthy population from Rio Grande do Sul (RS)/ Brazil and to estimate the prevalence of MCADD in the state. Methods: A total of 970 blood donors from RS/Brazil were included. DNA was extracted from blood samples in EDTA, using commercial kit, followed by PCR-RFLP using StyI restriction enzyme. Allele and genotypic frequencies were calculated considering Hardy-Weinberg Equilibrium. Results: c.985A>G was detected in heterozygosis in three subjects, resulting in a carrier and allele frequency of 1:323 individuals and 0.0015 respectively. MCADD prevalence due to c.985A>G in RS was estimated at 1:444,444 individuals. Conclusions: In this study, c.985A>G carrier frequency was found to be lower than for European countries. The low frequency observed may be due to several factors, such as the heterogeneity of the Brazilian population and the small sample size. Brazil is one of the most heterogeneous populations and this study highlights the need for expansion of FAOD investigation in the country.   2106 INVESTIGAÇÃO DA PREVALÊNCIA DA VARIANTE PATOGÊNICA C.1168G>A (ASSI), ASSOCIADA À CITRULINEMIA TIPO I, EM UMA POPULAÇÃO SAUDÁVEL DO RIO GRANDE DO SUL, BRASIL DA SILVA CMF; RANDON DN; DE BITENCOURT FH; SPERB-LUDWIG F; DOS SANTOS ML; MONTEIRO VCL; PORTELA LF; BECKER APP; VIANNA FSL; SCHWARTZ IVD HCPA - Hospital de Clínicas de Porto Alegre  Introdução: A citrulinemia tipo I (CTLNI) é um erro inato do metabolismo, causado por variantes patogênicas no gene ASSI, que expressa a argininosuccinato sintetase, uma enzima limitante no ciclo da ureia. A variante c.1168G>A (p.Gly390Arg), associada à forma neonatal grave da doença, representa até 62% dos alelos em pacientes europeus. Como a CTLNI não está incluída no Programa Nacional de Triagem Neonatal, estudos populacionais avaliando a frequência de variantes patogênicas tornam-se importantes para estimativas de prevalência e considerações de diagnóstico precoce. Objetivos: Avaliar a frequência da variante patogênica c.1168G>A (gene ASSI) em uma população saudável do Rio Grande do Sul (RS)/Brasil. Métodos: Mil indivíduos, doadores voluntários do Banco de Sangue do Hospital de Clínicas de Porto Alegre, foram incluídos. O DNA foi extraído a partir de sangue total em EDTA, utilizando kit comercial. A genotipagem foi realizada por PCR em tempo real, através do sistema Taqman. Resultado: A variante c.1168G>A esteve ausente em toda a população analisada. Conclusão: Este é o primeiro trabalho em avaliar a presença da variante c.1168G>A em uma população saudável do Brasil. Em concordância com a literatura, a ausência da variante reflete a raridade da CTLNI no RS.
