40a SEMANA CIENTÍFICA DO HOSPITAL DE CLÍNICAS DE PORTO ALEGRE Clin Biomed Res 2020; 40 (Supl.) 373 2375 HIPOPLASIA PULMONAR ENTRE RECÉM-NASCIDOS NO HOSPITAL DE CLÍNICAS DE PORTO ALEGRE DURANTE O ANO DE 2019: ESTUDO DE PREVALÊNCIA E ASSOCIAÇÃO COM OUTRAS MALFORMAÇÕES CONGÊNITAS BRUNA RUSCHEL; LETÍCIA ANTONIUK SEUS; PAULA CRISTINA DA COSTA; THANYSE DE OLIVEIRA SCHMALFUSS; JÚLIO CÉSAR LOGUERCIO LEITE; LAVÍNIA SCHULER FACCINI UFRGS -  Universidade Federal do Rio Grande do Sul  Introdução: A hipoplasia pulmonar (HP) é o desenvolvimento incompleto de um ou ambos os pulmões. Na maioria dos casos é secundária a outras malformações como: hérnia diafragmática, doenças renais (gerando oligodramnia), doenças neuromusculares. Há estudos que mostram ser a hérnia diafragmática a causa mais comum de HP, mas esse desenvolvimento incompleto dos pulmões também tem forte relação com disfunção renal, em decorrência do oligoidrâmnio. Em estudos retrospectivos, a incidência de hipoplasia pulmonar na população geral varia entre 9 a 11 a cada 10.000 nascidos vivos. Objetivos: Analisar a prevalência e malformações associadas a hipoplasia pulmonar entre os recém-nascidos (RNs) no Hospital de Clínicas de Porto Alegre (HCPA) no ano de 2019. Métodos: Foi realizado um estudo observacional transversal retrospectivo, através da análise de prontuários médicos do HCPA e do banco de dados do Programa de Monitoramento de Defeitos Congênitos do HCPA, Hospital colaborador do Estudo Colaborativo Latino-Americano de Malformações Congênitas (ECLAMC). Resultados: Em 2019 no HCPA, foram registrados 3.117 RNs vivos. Desses, dois apresentaram hipoplasia pulmonar, ambos associados a doença dos rins policísticos e anidrâmnio. Foi encontrado uma incidência de HP de 6,4 a cada 10.000 RNV. Quanto aos casos de hérnia diafragmática houve 5 casos, todos como malformação isolada. Conclusão: Os casos de HP entre os recém-nascidos no HCPA no ano de 2019 estavam associados a outra malformação (MF) congênita, concordando com a literatura, a qual mostra mais comum ser secundária a outra MF a vir de forma isolada. Contudo, nesse estudo, a relação da HP com hérnia diafragmática não foi mostrada. Ambos os casos de HP apresentavam doença dos rins policísticos associado a anidrâmnio. Estudos indicam que a maturação pulmonar depende da produção de líquido amniótico, o qual, após 15 semanas de gestação, é predominantemente produzido pelos rins fetais, dessa forma, oligo- ou anidrâmnios são suspeitos de disfunção renal e são associados ao desenvolvimento de hipoplasia pulmonar. A incidência de HP entre os RNV encontrado na literatura mostrou ser semelhante ao encontrado no HCPA durante o ano de 2019.   2420 SYSTEMS BIOLOGY APPROACHES REVEAL POTENTIAL PHENOTYPE MODIFIER GENES IN NEUROFIBROMATOSIS TYPE 1 LARISSA BRUSSA REIS; THAYNE WOYCINCK KOWALSKI; TIAGO FINGER ANDREIS; PATRICIA ASHTON-PROLLA; CLÉVIA ROSSET HCPA - Hospital de Clínicas de Porto Alegre  Introduction: NF1 is an atypical condition marked by varied symptoms that can range from mild to disabling and lethal. In general, symptoms can be grouped into large groups, according to disease-related ontologies. The groups involve manifestations in the central and peripheral nervous system, in metabolic/endocrine aspects and in bone development. However, because it is a syndrome with different pathways involved, the variants, and epigenetic changes already identified  in the NF1 gene are not yet sufficient to explain all the variability found in the disease. In this sense,  the concept of phenotype-modifier genes is extensively discussed in an attempt to explain this variability. Aim and Methods: To suggest potential NF1 modifier genes, we performed in silico analysis through systems biology tools using NF1 gene ontologies, its protein-protein interaction network, and its selected genes related to the NF1 phenotype and NF1 gene ontologies. Protein-protein interaction (PPI) networks were assembled with STRING tool v.10, and network statistics were performed in Cytoscape v.3.7.2, by using forward and reverse genetics strategies. We also performed heterogeneous networks analyses, comprising the phenotype ontologies selected, gene expression data, and the PPI network with PhenomeScape v.1.0.4 Finally, the hypothesized phenotype-modifier genes were verified by a random-walk mathematical model in R v.3.6.2 (RandomWalkRestartMH package). Results: The network statistics analyses combined with the forward and reverse genetics strategies, and the assembly of heterogeneous networks resulted in ten potential NF1 phenotype-modifier genes: AKT1, BRAF, EGFR, LIMK1, PAK1, PTEN, RAF1, SDC2, SMARCA4, and VCP. Mathematical models using the random-walk approach suggested SDC2 and VCP as the main candidate genes for phenotype-modifiers. Conclusions: We present here a previously unexplored systems biology strategy to investigate NF1 phenotype modifiers. The public availability of multi-omics datasets allows use of robust tools to generate complex networks including protein-protein interactions, differential expression data, and phenotypes, reinforced by mathematical models such as random-walk. Combining all these strategies, we found 10 candidate genes as potential NF1 phenotype modifiers.
