Ondanks een overvloed aan hersenstudies is één van de meest fundamentele basisprincipes van de hersenen – het feit dat we twee hersenhelften hebben met elk hun eigen specialisatie – nog altijd een mysterie. Deze hersenlateralisatie verschilt enigszins tussen mannen en vrouwen en daarom wordt gedacht dat het geslachtshormoon testosteron vóór de geboorte de hersenlateralisatie beïnvloedt als onderdeel van het proces waarbij ook andere sekseverschillen tot stand komen. De literatuur is hier echter niet éénduidig over, mogelijk doordat de noch rol van oestradiol (een ander geslachthormoon) noch de rol van hormonen in de puberteit (een andere belangrijke levensfase) is onderzocht. Ik onderzocht de invloed van testosteron en oestradiol vóór de geboorte en in de puberteit op de ontwikkeling van hersenlateralisatie. Daarnaast onderzocht ik de invloed van deze hormonen op de ontwikkeling van sekseverschillen in gedrag, namelijk genderidentiteit en rolgedrag. Hiervoor maakte ik gebruik van twee unieke data sets: 1) Een studie waarin we kinderen van vóór de geboorte (via een vruchtwaterpunctie bij de moeders) tot in de puberteit volgden. 2) Een studie naar de invloed van hormoontoediening op hersenlateralisatie bij adolescenten met genderdysforie (“transgenders”). Ik vond het volgende: 1. Hersenlateralisatie en genderontwikkeling zijn hele verschillende uitkomstmaten, maar we zagen overeenkomsten in de invloed van geslachtshormonen op beiden. 2. De invloed van hormonen in de puberteit hangt af van de blootstelling aan hormonen vóór de geboorte.3. Naast testosteron speelt ook oestradiol een belangrijke rol. 4. Het effect van geslachtshormonen hangt af van de ontwikkelingsfase, de cognitieve taak en het geslacht van de deelnemer.Sex hormones, like testosterone and estradiol, can affect the development of the brain and subsequently have essential influences on our behaviour. There are two major events during life when dramatic changes in the brain occur: before birth and in puberty. In the first part of the thesis, we investigated the influence of sex hormones on the asymmetrical specialization of cognitive functions of both halves of the brain (i.e. brain lateralization) based on two unique studies: 1) A longitudinal study in adolescents whose sex hormone levels were measured before birth (in the amniotic fluid of the mothers) . 2) A quasi-experimental study of the effects of hormone treatment in persons with gender dysphoria (who experience a mismatch between their sex assigned at birth and their gender identity). This offered us an experimental approach to study the effects of hormone administration in humans. In the second part of the thesis, we investigated the influence of sex hormones before birth and in puberty on gender role behaviour and gender identity from childhood to adolescence. Although the asymmetrical distribution of the brain and gender role behaviour and identity are very different output measures, there are several parallels in the effects sex hormones have. The main overarching conclusions are:1. Exposure to sex hormones before birth affects the influence of sex hormones in puberty. 2. Not only testosterone is important but estradiol plays a major role as well3. The effect of sex hormones depends on the phase of development, the cognitive task, and the sex of the participant
