Enzymologie is een zeer groot en interdisciplinair onderzoeksgebied. Enzymen zijn het onderwerp van intensief onderzoek om hun functies te onthullen in de stofwisseling en ook hun tekortkomingen die tot verschillende ziekten kunnen leiden. Daarom zijn enzymen vaak het doelwit in geneeskundig en farmaceutisch onderzoek. Enzymen spelen niet alleen in de geneeskunde een belangrijke rol, maar ook bijvoorbeeld in de voedingsindustrie. Een interessant feit is dat belangrijke enzymen uit gist sinds de oudheid zijn gebruikt voor de productie van wijn, kaas en brood. Aan het eind van de 20e eeuw brak voor de enzymologie een nieuw tijdperk aan: ‘protein engineering’. Dit werd veroorzaakt door spectaculaire ontwikkelingen omtrent het bepalen van eiwitstructuren , genmanipulatie en sequencing technieken. Ook door de toenemende computerkracht en nieuwe software waren de gevolgen van mutaties in enzymen vooraf beter en makkelijker te voorspellen. Dit opende de weg naar het ontwerpen van enzymen voor industriële doeleinden en tegenwoordig worden veel enzymen daar succesvol toegepast. Er wordt nu nog steeds veel onderzoek gedaan om dit gebied verder te ontwikkelen. Dit proefschrift richt zich op de structuur, de activiteit en het mechanisme van een aantal oxidatieve enzymen. Ze zijn allemaal afhankelijk van een flavine cofactor, die een centrale rol speelt in de reacties die worden gekatalyseerd.Enzymology is a very large and interdisciplinary research field and enzymes have been the subject of intense research in order to unveil their functions in metabolism and their shortcomings leading to different diseases. Therefore, enzymes are a very common target in medicine and pharmaceutical industry research. What is interesting to note, they have been used since ancient times for the production of wine, cheese and bread through the use of yeast. At the end of the 20th century enzymology entered a new era of protein engineering. This was due to a spectacular development in determining protein structures, gene manipulation and sequencing, as well as in computing power and software that can predict the impact of mutations introduced to an enzyme. This also opened the road to enzyme design for industrial purposes and nowadays many enzymes have been successfully applied and still a lot of research effort is spent on further development in this field. This thesis was intended to study the structure, activity and mechanism of several oxidative enzymes. They all depend on a flavin cofactor which plays a pivotal role in the reactions they catalyse

Kopacz  1. The most intriguing question regarding putrescine oxidases is the reason why they bind ADP (chapter 3).  2. It is well known that flavoproteins show amazing biochemical versatility, which hinders the design of generic approaches for their engineering (chapter 3 and 4).  3. The bicovalent flavin binding sequence motif will facilitate the discovery of new interesting biocatalysts (chapter 4).  4. Although a methyl group does not normally exchange protons with solvent, a proper electron sink may in some cases promote such exchange (chapter 5).  5. “The right understanding of any matter and a misunderstanding of the same matter do not wholly exclude each other” (“The Trial”, Franz Kafka). The absurdity described in the novel is a very good representation of what bursary PhD students have to deal with.  6. Einstein’s theory of relativity can be somewhat exemplified by the Dutch railway. The travel distance  from Groningen to Amsterdam seems not the same as that from Amsterdam to Groningen.  7. The Netherlands is considered as one of the most developed and richest countries in the world, but this is not reflected in the food variety readily found in the supermarkets.
