Het onderzoek van Rosalina Wisastra was gericht op de ontwikkeling van de chemiegebaseerde methodes die kunnen worden gebruikt om een beter begrip van enzymactiviteit in ontstekingsprocessen te verkrijgen via de NF-κB pathway. In het eerste deel van haar proefschrift beschrijft Wisastra de ontwikkeling van nieuwe modulatoren voor lipoxygenase (LOX) enzymactiviteit. Enzymkinetische studies aan deze salicylaatgebaseerde modulatoren laten zien dat zij de lipoxygenaseactiviteit reguleren door te binden aan een allosterische regulerende bindingsplaats in lipoxygenses. Het tweede deel beschrijft een grondig onderzoek van isothiazolonhetercycli. Dit zijn thiolaatreactieve functionaliteiten die gebruikt kunnen worden voor de ontwikkeling van nieuwe moleculen voor activiteits-gebaseerde eiwitprofilering (ABPP) van enzymen die een rol spelen bij ontstekingen. Een nieuw opgehelderde kristalstructuur van de isothiazolonheterocyclus laat een aromatisch karakter zien, dat stabiliteit biedt. In tegenstelling daartoe maakt de zwakke zwavel-stikstofbinding een snelle en selectieve reactie met thiolaten tot een sterke disulfidebinding mogelijk. Het laatste gedeelte beschrijft de ontwikkeling van een nieuwe eenvoudige kleuringsmethode voor oxidatieve stress in weefselsecties met fluorescentiemicroscopie. Deze nieuwe methode is selectief voor tyrosinenitrering, een oxidatieve eiwitmodificatie die vaak wordt waargenomen bij ontstekingen. Deze nieuwe methode is gebaseerd op de chemische omzetting van nitrotyrosine in een fluorofoor onder milde omstandigheden. Deze kleuringsmethode bleek zeer efficiënte voor secties van ontstoken weefsel en kan ook gebruikt worden als alternatieve kleuringsmethode voor immunoblotting op PVDF-membranen
