Bodem bacteriën van de genus Streptomyces zijn biotechnologisch zeer interessant omdat ze één van de belangrijkste bronnen van natuurlijke antibiotica zijn die tegenwoordig gebruikt worden in de geneeskunde. Deze bacteriën communiceren met hun soortgenoten en coördineren aldus de synthese van antibiotica om het efficiënter te maken. Zij communiceren door kleine chemische signaalmoleculen te gebruiken. In Streptomyces coelicolor, het onderwerp van onze studie, die diverse antibiotica produceert, worden deze moleculen SCBs (Streptomyces coelicolor Butyrolactone) genoemd. Eerder onderzoek aan het SCB signaalsysteem in deze bacterie, heeft zijn relevantie in de controle van antibiotica productie aangetoond, en ook de grote complexiteit van dit systeem dat nog niet helemaal begrepen wordt. In dit doctoraat proefschrift hebben wij de rollen van de eiwitten die betrokken zijn in dit communicatie systeem bestudeerd. We hebben nieuwe essentiële eiwitten geïdentificeerd en gekarakteriseerd die betrokken zijn bij de synthese van SCB signaalmoleculen, en nieuwe regulerende factoren betrokken bij het juiste functioneren van dit systeem. De hier gepresenteerde informatie draagt bij tot een beter begrip van zulke complexe reguleringsmechanismen en van de regulering van antibiotica productie in deze bacteriën. Dit is van groot belang voor het ontwikkelen van nieuwe strategiën om de productie van antibiotica te verhogen door biosynthetische routes van antibiotica te manipuleren en voorziet in belangrijke instrumenten om nieuwe natuurlijke stoffen met antimicrobiologische activiteit te ontdekken.Soil bacteria of the genus Streptomyces are of great biotechnological interest since they are one of the main sources of the natural antibiotics used nowadays in medicine. These bacteria communicate with other members in their population to coordinate the synthesis of antibiotics to make it more efficient. For this purpose they use small chemical signalling molecules. In Streptomyces coelicolor, the target of our study, and producer of several antibiotics, these molecules are called SCBs (Streptomyces coelicolor Butyrolactone). Previous research on the SCB signalling system in this bacterium showed its relevance in the control of antibiotic production and the large complexity of this system that is yet not fully understood. In this PhD thesis the roles of the proteins involved in this communication system in S. coelicolor have been extensively studied. We have identified and characterised additional essential proteins involved in the synthesis of the SCB signalling molecules and described new regulatory factors that govern the functioning of this system. The information presented here contributes to a better understanding of such a complex regulatory mechanism and of the regulation of antibiotic production in these bacteria. This is of great relevance for the design of new strategies to increase the production of antibiotics by manipulating antibiotic biosynthetic pathways and provides important tools for the discovery of new natural compounds with antimicrobial activities
