BackgroundAllergen immunotherapy only has a moderate efficacy, both inhumans and animals. During sublingual immunotherapy, the oralmucosa is an easily accessible, immunologically active tissuewhich could provide the opportunity to modify the local immuneresponse. In humans and mice, oral dendritic cells1 and thedraining lymph nodes of the oral mucosa2 are known to play animportant role in the promotion of tolerance during therapy. Skinand lung epithelial cells can influence the co-stimulatory signal ofantigen-presenting cells3. Whether oral epithelial cells are capableof immunologically recognizing allergens and could be modulatedin playing a tolerance-promoting role has not been specificallyinvestigated.Objectives of the study1. To look at the influence of allergens on oral epithelial cells’cytokine secretion2. To identify substances capable of suppressing thepro-inflammatory immune response in oral epithelial cellsMethodsA cell line of canine oral keratinocytes was incubated with allergenextracts (Dermatophagoides farinae, D. pteronyssinus andAlternaria alternata), different toll-like receptor ligands, caninerecombinant cytokines and vitamins (9-cis-retinoic acid andcalcitriol). After 24 hours incubation, cytokine production (IL-8and TGF-β1) was measured with ELISA.Results1. Influence of allergens on oral epithelial cells’ IL-8 and TGF-β1 cytokine secretion2. Effect of TLRL, cytokines and vitamins on IL-8 and TGF-β1 cytokine secretion in oral epithelial cellsConclusion Just as in skin keratinocytes, house dust mite antigens seem tobe capable to induce pro-inflammatory signals in oral epithelialcells. Calcitriol on the other hand seems to downregulatecytokine production in this cell type. The effect of allergens andcalcitriol on oral epithelial cytokines that are specificallyinvolved in Th2 immunity should be further investigated. Abetter understanding of the different substances that canmodulate immune responses at the level of the epithelium willallow the selection of specific adjuvants that could enhance theefficacy of local allergen immunotherapy.References1Allam et al.
