With regard to this fact, the underlying physiological and molecular mechanisms of weight gain are of great interest. We hypothesized that in cats body fat content and also a reduction of the body fat content has an effect on transcriptome profile in white adipose and skeletal muscle tissue. Differences of this were expected in cats lean versus overweight cats.  Animals, materials and methods: Six female cats (3 body condition score (BCS) <5.5 = lean), 3 BCS>6/9 = overweight [2]) were used. Overweight cats underwent a weight-loss program before the experiment. At the first sampling time (T1) all cats were lean (BCS <5.5), white adipose and muscle tissue biopsies were taken under anaesthesia. Thereafter all cats were fed ad libitum and three cats gained weight again (BCS ≥ 6=group GO) after weight loss program), the remaining three cats stayed lean at each time of the experiment (BCS <5.5 GL). When the GO cats were overweight a second tissue collection was done (T2). The experiments were approved by Swiss veterinary authorities (ZH128/2014). The cats were fed with a commercial dry food (Classic Chicken, Biomill SA, Herzogenbuchsee, CH). At T1 and T2 blood samples were taken and body fat content was determined using Dual energy X-ray absorptiometry (DEXA) under sedation and in sternal recumbency (Hologic QD A 85508 discovery machine and QDR APEX system software version 3.3, 35 Crosby Drive, Bedford, Ma, USA). Blood glucose was analysed immediately with a blood glucose meter (Ascencia EliteTM, Bayer Inc. Healthcare Division, Toronto, Canada). RNA was extracted from all tissue samples using the Trizol protocol (Invitrogen, Carlsbad, CA, USA), quality and quantity was tested. Good quality samples were analysed at Ramaciotti Centre for Genomics (University of New South Wales). Individual libraries were pooled and next-generation sequences generated. Correlations of BCS with body fat content, resp.or blood glucose was performed using SPSS (IBM Corp. Released 2017. IBM SPSS Statistics for Windows, Version 25.0. Armonk, NY: IBM Corp.). RNAseq Data analysis was conducted using R (Version 4.0.3). Results and Discussion: Lean cats had a mean body fat (BF) content of 12% (SD ± 4.00) The BCS of group GO were at T1=5.2±0.3 and of GL at T1 5.0±0.14 and at T2 5.0±0.04). BW o GL at T1 was 3.3±0.4kg, at T2 3.5±0.3 and GO at T1 3.6±0.5 and T2 4.7±0. In GO cats (BCS=6.8±0.3) the mean BF value was 27% (SD ± 5.32). Blood glucose was with lean 4.7 mmol/l (SD ± 0.51) and overweight (BCS ≥ 6) 4.6 mmol/l (SD ± 0.26) always in a physiological range. In the present experiment, there was no clear correlation between transcriptome and group in the different tissues. Also between different time points, only small differences in gene regulation could be seen (GL at T2 over GL at T1 muscle tissue: 7 genes downregulated and 33 genes upregulated GO at T1 over GO at T2, 3 genes downregulated, 4 genes upregulated). In adipose tissue, the difference was even smaller. The missing relevant gene reaction could be explained by the fact that the cats were overweight, but not heavily obese and that all physiological processes like e.g. glucose metabolism were in a physiological range. Another explanation would be that the investigated tissues are not the first to react. Liver tissue could have led to different results.
