Multicomponent reactions (MCRs) are one of most powerful tools to synthesize novel scaffold compounds for drug discovery. 2. New synthetic routes for rigid nitrogen-containing polyheterocycles using sustainable, atom-economical and efficient (=short) synthetic routes are highly desired. 3. Multicomponent reaction (MCR)-based compound libraries combined with virtual screening are useful strategies for identifying MIF2 tautomerase inhibitors. 4. X-ray crystallography combined with a multicomponent reaction (MCR) approach could be a very promising approach to develop small-molecule inhibitors of P. falciparum ATCase.  5. The multicomponent reaction (MCR) approach can not only generate antimalarial compounds, but also develop small molecules targeting human ATCase as anticancer drugs. 6. Don’t aim for success if you want it; just do what you love and believe in, and it will come naturally.  7. Scientists must grasp some universal features that can be expressed by precise formulas in the vast and complex facts of experience, thus exploring the universal principles of nature. ----- Albert Einstein
