ANDERSON University of Wisconsin, Madison, Wisconsin,-and Central High School, Norwood, Minnesota One limitation in studying human blood groups is the scarce sup-ply of certain typing fluids (reagents). Even reagents that are readily available are usually from different individuals and often show varia-tion in specificity and strength. Therefore, a source of large quantities of strong and specific reagents from a single individual would be use-ful, especially for genetic studies. Cattle are readily available and are a good source of large amounts of sera. The naturally occurring anti-bodies for the J factor of cattle also react with human A cells. Such normal anti-J sera can be used as specific human A reagents at high dilutions (1/64-1/256) without absorption and at dilutions as high as 1/256 even after absorption with human O cells. Also, because of seasonal variation in strength of anti-J ( ergo anti-A) normal sera obtained from cattle during summer months consistently show high titers. Cattle lacking anti-J and possessing soluble J substance pro-vide an excellent source of human anti-O(H) antibodies. We found that about 30% of the I-positive Holstein cattle had naturally occur-ring antibodies from which anti-O(H) could be prepared. We also found that cattle can be used to produce immune sera against human cells. We have prepared antisera in 18 cattle against the cells of six human donors. Each of three cows immunized with human M cells produced sera from which potent specific anti-M reagents have been prepared. These reagents were tested with more than 100 cells of known type and have reacted strongly and specifically with cells of type Mand MN. Preliminary analyses of these cattle anti-human sera indicate that at least three "new" specificities of human cells have been detected. 291
