Research on Multicolor Melting Curve Analysis-Based Approaches for Gene Mutations Detection

Abstract

作为一种新型的均相实时PCR检测技术,多色探针熔解曲线技术具有简便快速、特异性好、灵敏度高、结果准确可靠、检测成本低等优点,因此被用于遗传、肿瘤及微生物等领域的检测。然而,该技术的检测对象主要是点突变(包括错义突变、移码突变、小片段缺失或插入突变等),目前尚无报道将该技术用于大片段缺失突变的检测和高GC含量的长片段PCR扩增子的检测。因此,本论文围绕多色探针熔解曲线技术在不同类型基因变异中的检测进行研究,旨在进一步证明该技术的灵活性及适用性。 第一章为绪论部分,首先简要介绍了基因变异与人类疾病之间的关系以及基因检测技术的发展,然后系统介绍了探针熔解曲线分析技术的发展、特点以及目前适合的荧光探...As a novel homogeneous real-time PCR, multicolor melting curve analysis is cost-effective, easy-to-use, accurate, high specificity and sensitivity, therefore it is widely applied in the detection of genetic diseases, tumor and microbiology.
