Study on the influence of human hepatocellular carcinoma multidrug resistance by MAPK signal pathway

Abstract

目的: 1.化疗药物诱导建立多株人肝癌多药耐药模型,鉴定其耐药性,比较亲本株与耐药株生物学特点的差异。 2.检测人肝癌细胞亲本株及耐药株中SP细胞比例,探索耐药细胞与SP细胞的联系。 3.检测人肝癌多药耐药细胞模型中MAPK表达情况,探索人肝癌细胞株中MAPK信号通路与多药耐药的关系。 4.检测人肝癌组织中MAPK与MDR1表达情况,探索人肝癌组织MAPK信号通路与多药耐药的关系,以及MAPK在肝癌组织中的表达情况。 方法: 1.采用ADM对HepG2、SMMC-7721、BEL-7402等三株人肝癌细胞株进行大剂量冲击和小剂量缓升两种方法诱导。采用生物发光法及MTT法进行药敏实验...Objective 1.To induce poly multidrug resistance (MDR) cell models from human hepatocellular carcinoma (HCC) cell lines by chemotherapy drugs, and characterize their resistance ability.
