阿尔茨海默病（Alzheimer'sdisease，AD）是最常见的神经退行性疾病，神经元外过度产生的淀粉样蛋白Aβ（Amyloidβ）异常堆积形成的老年斑（SenilePlaque，SP）是其一个重要的病理特征。当前的研究已经表明γ-分泌酶的活性对于Aβ的产生非常重要，APP在β-分泌酶和γ-分泌酶共同作用下水解产生Aβ。早老素蛋白-1（presenilin1,PS1）是γ-分泌酶的重要成份。PS1水解片断聚合形成二聚体，与其它的γ-分泌酶成份相互作用形成多功能复合体，从而调节后者对APP的切割活性。研究也已经表明PS1蛋白突变可影响γ-分泌酶的活性及APP的代谢，并显著减少Aβ40和Aβ4...Alzheimer disease (AD) is a neurodegenerative disorder of the central nervous system.
