Studies on the Chiral Synthesis of 3-Hydroxypiperidines and the Asymmetric α-Hydroxyalkylation of β-Hydroxypyrrolidine

Abstract

摘要哌啶类生物碱和吡咯烷类生物碱是两类重要的生物碱,由于大多具有重要的生理活性,所以其不对称合成一直吸引人们的兴趣。本论文的目的是拓展本实验室基于手性源的不对称合成方法学研究,主要研究从天然的L-谷氨酸出发不对称合成3-羟基哌啶类化合物的方法。同时,在实验室已有工作基础上,研究手性吡咯烷碳负离子的形成及反应。取得的主要结果如下:一、从L-谷氨酸衍生物I出发,经酸催化开环、叔丁基二甲基硅基保护、酯基还原三步,高产率地合成了重要的手性砌块II,三步总产率82%。二、从手性合成砌块II出发,探索了反式-3-羟基哌啶-2-羧酸III的合成方法,最终完成了其类似物反式-3-羟基哌啶-2-酰胺IV的合成。...ABSTRACT Piperidine and pyrrolidine alkaloids are two important class of alkaloids.
