Denne rapport undersøger vandopløseligheden af to stoffer: usninsyre og dens derivat, kaldet dimer. Usninsyre er grundet dens anticancer egenskab et stof, der ønskes undersøgt som en mulig behandling mod kræft. Dog har usninsyre vist sig at være alvorligt levertoksisk, hvor det ikke vides, om dimer har samme egenskaber. Det fastlås i rapporten, at dimer er mere opløseligt end usninsyre. Der undersøges, hvorvidt begge stoffers vandopløselighed kan øges ved brugen af cyklodextriner, som hyppigt bruges i medicinalindustrien til selv samme formål. På trods af udførelsen af gentagende forsøg, kan vi ikke bekræfte denne hypotese udelukkende på baggrund af vores resultater. Cyklodextriner gør usninsyre og dimer mindre vandopløseligt i vores forsøg, hvilket tyder på en fejl i metoden. Efter dette udføres der forsøg i Caco-2 celler, som er en kræftcellelinje fra en adenokarcinom i tarmen, for at se hvorvidt begge stoffer påvirker celleviabiliteten med og uden brugen af cyklodextriner. Vi konkluderer ud fra vores og lignende forsøg, at usninsyre har en cytotoksisk effekt på maligne celler. Vores forsøg viser ligeledes samme effekt med brugen af dimer, dog ikke i lige så høj grad som med usninsyre. Dette åbner op for en spændende tilgang til nye undersøgelser med stoffet dimer.This paper studies the solubility of two drugs: usnic acid and its derivate, called dimer. Usnic acid is, because of its anticancer qualities, a drug which should be studied as a possible cancer treatment. However, usnic acid has shown to be seriously hepatotoxic, and it is not known, if dimer has the same characteristics. In this paper, it is established that dimer is more soluble than usnic acid. We examine whether the solubility of usnic acid and dimer can be improved using cyclodextrins, which is commonly used in the pharmaceutical industry for this purpose. Despite the repetition of experiments, we could not confirm this hypothesis. Our experimental results concluded that cyclodextrins reduced the solubility of usnic acid and dimer, which points toward a methodological flaw. Afterwards, we conducted experiments to examine how both drugs, with and without cyclodextrins, affect the cell viability of an adenocarcinoma cell line (Caco-2 cells). Based on our experiments as well as other similar experiments, we can conclude that usnic acid has cytotoxic qualities on malignant cells. Likewise, our experiments show the same cytotoxic activity by dimer, although not as much as with usnic acid. This creates a new and exciting approach for new studies on dimer
