En vedvarende problematik i udviklingen af nye lægemidler i den farmaceutiske industri er den dårlige opløselighed af ellers sundhedsfremmede molekyler. Den lave vandopløselighed fører til dårlig absorption af lægemidlet, hvilket vil medføre lav biotilgængelighed.Opløseligheden af et molekyle afhænger af dens struktur. Der er flere teknikker til forbedring af opløseligheden. Den teknik, som vi benytter os af i vores projekt, er kompleksdannelse med cyklodextriner, som forbedrer vandopløseligheden og biotilgængeligheden af dårligt vandopløselige molekyler. Vi fokuserer på molekylerne usninsyre og dens kemisk modificerede version, dimer. Komplekser dannes ved indsættelse af det ikke-polære molekyle (gæst molekyle) i hulrummet af et andet molekyle (vært molekyle).Formålet med dette projekt er at undersøge molekylernes opløselighed og effekten af cyklodextriner på de to molekyler; usninsyre og dimer. Herefter vil vi undersøge effekten af de to cyklodextrin komplekser på celleviabilitet i cellelinjen Caco-2.Konklusionen er at vi kan forbedre opløseligheden af molekylerne ved hjælp af cyklodextriner. Begge molekyler hæmmer celleviabiliteten på Caco-2 cellelinjen.One inherent problem with developing new drugs in the pharmaceutical industry is the poor solubility of the compounds. Low aqueous solubility leads to defective absorption of the drug, and this will cause low bioavailability.The solubility of a compound depends on its structure. There are several techniques to enhance solubility. The technique that we have used in our project is inclusion complex formation with cyclodextrins, which improves aqueous solubility and bioavailability of poorly water-soluble molecules. We focus on the molecules usnic acid and a chemically modified version, dimer. Inclusion complexes are formed by the insertion of the nonpolar molecule (guest) into the cavity of another molecule (host).The aim of this project is to investigate the solubility of the molecules and the effect of cyclodextrins on the two solubility of the two compounds; usnic acid and dimer. Further, we investigate the effect of these two cyclodextrins complexes on cell viability in the cell line Caco- 2.We found that we can improve the solubility of the molecules with the assistance of cyclodextrins. Both molecules function as inhibitors on the cell viability of the Caco-2 cell line
