Conventional cancer medicine often has short-term therapeutic effects, which lead to drug resistance in some cases. Pharmaceutical researchers' attempts are focused on the synthesis of pharmaceutical compounds with selective mechanisms of action and less adverse effects. Methods: Dihydro-pyranopyran and Dihydro-pyranochromene derivatives were used for in-vitro studies after synthesis and identification by researchers of Kerman School of Pharmacy. The cytotoxic effects of these compound were determined using MTT Proliferation Assay. Thus, 10000 cells were cultured in each well of a 96-well microplate. The cells were treated with different concentrations of the synthetic compounds. In each experiment, control groups of the cells and the solvent were also considered. MTT and then DMSO solution was added to each well of the microplate 24 hours after treatment and the absorption was measured at 570 nm after solubilizing the formed formazan crystals. Finally, dose-response curves were plotted and the concentrations that inhibited the cell growth by 50% (IC50), were calculated for each compound. Results: According to the calculations, the best effect (lowest IC50) on the MCF7 cell line, was related to the B5 compound (102 µg/ml).This compound also had the lowest IC50 (164 µg/ml) on the 3T3 cell line. Conclusion: The B5 compound showed a more cytotoxic effect on the tested cell lines probably due to electron-withdrawing group (NO2) on its side ring. This compound can be considered in future studies.
