Among many Cordyceps s.l., Ophiocordyceps sinensis or Cordyceps militaris, known as “To-chu-ka-so” in Japan, have been extensively examined regarding their medicinal use. Several biological activities and pharmacological effects of Cordyceps s.l. have been reported, including liver function improvement, anti-cancer, anti-oxidation, anti-aging, and anti-diabetic activities. It was also suggested that Cordyceps s.l. can be used to support conventional cancer therapies, such as surgery, radiation therapy, and chemotherapy to reduce their side effects. Regarding its anti-cancer activity, Cordyceps s.l. induce apoptosis or cell cycle arrest in cancer cells, and reduce metastasis in experimental animal models. In addition, inhibition of the nuclear factor kappa B (NF-κB) signaling pathway or the down-regulation of tumor-promoting inflammatory cytokine expression in cancer cells may be the molecular mechanism of the anti-cancer effects of Cordyceps s.l.. Inflammatory genes are the key players in tumor progression and metastasis, and are known to be controlled by transcription factors such as NF-κB or signal transducer and activator of transcription 3 (STAT3). As the inflammatory cytokine IL-6 is a potent activator of STAT3 and the main target gene of NF-κB, the NFκB-STAT3-IL-6 inflammatory pathway is considered an attractive pharmacological target to control the pathogenesis of many types of tumors. Although more than 600 species of Cordyceps s.l. have been identified, most have not been well explored regarding their potential for medicinal use.  In this study, the profiles of constituents of ten different species of Ophiocordyceps, which is an unexplored species of Cordyceps s.l., were analyzed and their anti-tumor effects were examined further by targeting the NFκB-STAT3-IL-6 inflammatory pathway. Furthermore, bioassay-guided investigation of the extract of Ophiocordyceps gracilioides using 4T1 breast cancer cell lines expressing a luciferase  reporter gene of NF-ĸB or STAT3 led to the isolation of the active components with anti-metastatic activity in O. gracilioides.  1. Identification of Ophiocordyceps gracilioides  by its anti-tumor effects through targeting the NFκB-STAT3-IL-6 inflammatory pathway (参考文献 1)  The profiles of constituents of ten different species of Ophiocordyceps, which is an unexplored species of Cordyceps s.l., were analyzed and their anti-tumor effects were further examined. Although all Ophiocordyceps samples exhibited similar peak patterns, O. gracilioides had a distinct constituent profile from the other samples. Furthermore, O. gracilioides was the most active in suppressing the transcriptional activities of both N F-κB and STAT3, and the production of IL-6 from breast cancer cells. This study demonstrated that O. gracilioides is a relatively unexplored Cordyceps s.l. that may have medicinal potential to inhibit the NFκB-STAT3-IL-6 inflammatory pathway in cancer.   2. Anti-metastatic effects of ergosterol peroxide from the entomopathogenic fungus Ophiocordyceps gracilioides  on 4T1 breast cancer cells（参考文献 2）   The bioactive compounds ergosterol peroxide and ergosterol from the methanolic extract of O. gracilioides mycelia were identified related to its anti-cancer effects by targeting the NF-ĸB and STAT3 inflammatory pathways. Using gene-reporter assays, I demonstrated that ergosterol peroxide markedly inhibits both NF-ĸB and STAT3 activity in 4T1 cells, whereas ergosterol had limited effect. Consistent with their effects on NF-ĸB and STAT3 activity, ergosterol peroxide, but not ergosterol, exerted anti-proliferative effects on 4T1 cells. Furthermore, ergosterol peroxide significant inhibited both the migration and invasion of 4T1 cells in vitro, and pre-treatment of 4T1 cells with ergosterol peroxide significantly inhibited the formation of experimental lung metastases in vivo. Collectively, I demonstrated that ergosterol and ergosterol peroxide can be isolated from O. gracilioides mycelia, and further identified ergosterol peroxide as an active constituent of its anti-metastatic effects through the inhibition of NF-ĸB and STAT3 inflammatory pathways in 4T1 breast cancer cells.     参考文献： 1. Med., 1-9, Online published (Apr 27 th). doi: 10.1007/s11418-021-01520-2  様式７ 学位論文審査の要旨  報告番号   富医薬博甲第   号    富医薬博乙第   号   氏   名   Min-Kyoung Shin  審査委員               職 名              氏    名  （主査）     教 授       櫻井  宏明              （副査）     教 授       小松  かつ子              （副査）     教 授       早川  芳弘              （論文題目）（英語の場合は和訳，日本語の場合は英訳を付記すること｡英語(訳 )は，最初の文字のみを大文字で表記し，他は小文字で表記すること（ただし，学名等を除く）。）  Identification of Ophiocordyceps gracilioides as an antitumor natural drug resource（抗腫瘍性天然薬物資源としてのOphiocordyceps gracilioides の同定）  （判定）  合 格  （論文審査の要旨）（２頁以内）  冬虫夏草は昆虫類に寄生した菌糸から子実体を作る子囊菌類の一種で、貴重な生薬資源として知られている。これまでは抗がん作用などについて Ophiocordyceps sinensis やCordyceps militaris（サナギタケ）を中心に研究されているが、その他の種では希少性や菌株の分離・培養の難しさ等から研究例は少ない。本博士論文では富山県森林研究所で所有している冬虫夏草菌株ライブラリについて、それらの薬理学的特徴を明らかにすることで新たな薬用資源としての有用性を見出した研究成果が報告されている。特に、がん細胞において腫瘍促進的に働くことが知られている転写因子 Nuclear factor-kappa B（NF-B）とSignal transducer and activator of transcription 3 (STAT3)に着目し、冬虫夏草菌のなかでもOphiocordyceps 属について探索研究している。 まず第 1 章では、10 種類の冬虫夏草菌ライブラリの薬理活性について、マウス乳がん4T1 細胞に対する抗腫瘍作用、ならびに NF-B と STAT3 抑制作用を指標に比較検討している。その結果、Ophiocordyceps gracilioides（ウスイロタンポタケ、以下 O. gracilioides）が、がん細胞増殖抑制作用、ならびに NF-B と STAT3 の抑制作用、IL-6 産生抑制作用を最も強く示す事を見出した。冬虫夏草菌株の成分プロファイリングでは、O. gracilioides は他の菌株と比較して異なる成分を保有していることが示唆された。  さらに第 2 章では、O. gracilioides の抗がん作用を担う活性成分の同定を目標にした探索研究の成果が報告されている。その結果、O. gracilioides のメタノール抽出物から、4T1細胞の NF-κB と STAT3 の両方を阻害する活性化合物として、ergosterol peroxide およびergosterol を同定している。さらに 4T1 細胞における NF-κB および STAT3 活性抑制作用、ならびに増殖抑制作用は ergosterol に対して ergosterol peroxide がより強い効果を示した。これらの結果に加えて、ergosterol peroxide は in vitro での 4T1 細胞の浸潤・移動能を抑制し、さらに ergosterol peroxide による 4T1 細胞の前処理は、in vivo 実験的肺転移モデルでの肺転移形成を有意に阻害した。以上の結果は、O. gracilioides の新たな薬用資源としての重要性を明らかにし、加えて ergosterol peroxide は O. gracilioides の抗腫瘍活性を担う重要な薬理活性成分であることを初めて示した大変重要な研究成果であると評価できる。 主査及び副査は、申請者 Min-Kyoung Shin 氏の本学位論文の内容を精査するとともに面接審査を行い、Min-Kyoung Shin 氏が博士（薬科学）の学位を受けるに十分に値すると判断した。  (学位論文のもとになる論文 著者名,論文題目,掲載誌名,巻,最初の頁と最後の頁,年を記載)                1. 著者名：Shin, M.-K., Sasaki, F., Ki, D.-W., Win, N.N., Morita, H., Hayakawa, Y.  論文題目：Identification of Ophiocordyceps gracilioides  by its anti-tumor effects through targeting the NFĸB-STAT3-IL-6 inflammatory pathway.  雑誌名・巻・頁・年：Biol.
