先端ライフサイエンス研究拠点 Frontier Research Core for Life Sciences 天然物創薬分野 Division of Natural Drug Discovery  特命助教 アワレ スレス Suresh Awale 研究員 上田 純也  Jun-ya Ueda  ◆ 研究概要  我々の分野は，主として，腫瘍微小環境を標的とする天然抗がん物質の探索を目的に研究を進めている。一般的ながん細胞は無秩序かつ急速に増殖するが，腫瘍血管系が脆弱で無秩序に形成されるために栄養や酸素の欠乏した環境にさらされることになる．しかしながら，がん細胞は低栄養・低酸素といった極限状態におかれると，エネルギー代謝を変えることで生存する特有の耐性機構を示す．特に，PANC-1 のようなヒト膵臓がん細胞はこのような耐性を獲得しており，低栄養・低酸素といった厳しい環境下においても長期間の生存が可能となっている．したがって，がん細胞の栄養飢餓耐性を解除する化合物（anti-austerity agent）は新たな抗がん剤探索の標的と考えられる．ほとんどの膵臓がん患者は速やかに転移を起こし，短期間で死に至る．これまで膵臓がんに対する有効な薬はなく，従来の抗がん剤に対しては耐性を示す．それゆえ，栄養飢餓耐性を標的とする天然抗がん物質の探索が，治療戦略において重要な研究課題の一つであると考えられる．この目的を達成するために，1) 各地の様々な植物資源の膵臓がん細胞株に対する抗がん活性スクリーニング，2) 生理活性を指標とした活性物質の探索，3) 定量的メタボロミクス解析を利用した抗がん剤の作用機構の解明，4) 動物（マウス）における抗腫瘍活性評価を進めている．  ◆ 原 著 1)  Awale S., Linn T.Z., Li F., Tezuka Y., Myint A., Tomida A., Yamori T., Esumi H., and Kadota S. : Identification of chrysoplenetin from Vitex negundo as a potential cytotoxic agent against PANC-1 and a panel of 39 human cancer cell lines (JFCR-39).
