Ironing out the details: Untangling dietary iron and genetic background in diabetes
The search for genetic risk factors in type-II diabetes has been hindered by a failure to consider dietary variables. Dietary nutrients impact metabolic disease risk and severity and are essential to maintaining metabolic health. Genetic variation between individuals confers differences in metabolism, which directly impacts response to diet. Most studies attempting to identify genetic risk factors in disease fail to incorporate dietary components, and thus are ill-equipped to capture the breadth of the genome’s impact on metabolism. Understanding how genetic background interacts with nutrients holds the key to predicting and preventing metabolic diseases through the implementation of personalized nutrition. Dysregulation of iron homeostasis is associated with type-II diabetes, but the link between dietary iron and metabolic dysfunction is poorly defined. High iron burden in adipose tissue induces insulin resistance, but the mechanisms underlying adipose iron accumulation remain unknown. Hepcidin controls dietary iron absorption and distribution in metabolic tissues, but it is unknown whether genetic variation influencing hepcidin expression modifies susceptibility to dietary iron-induced insulin resistance. This review highlights discoveries concerning the axis of iron homeostasis and adipose function and suggests that genetic variation underlying dietary iron metabolism is an understudied component of metabolic disease
Does diet mediate associations of volume and bouts of sedentary time with cardiometabolic health indicators in adolescents?
Objective: Examine the mediating role of diet in the relationship between volume and duration of sedentary time with cardiometabolic health in adolescents. Methods: Adolescents (12-19 years) participating in the 2003/04 and 2005/06 U.S. National Health and Nutrition Examination Survey (NHANES) were examined. Cardiometabolic health indicators were body mass index z-scores (zBMI) (n 5 1,797) and metabolic syndrome (MetS) (n 5 812). An ActiGraph hip-worn accelerometer was used to derive total sedentary time and usual sedentary bout duration. Dietary intake was assessed using two 24-hour dietary recalls. Mediation analyses were conducted to examine five dietary mediators [total energy intake, discretionary foods, sugar-sweetened beverages (SSB), fruits and vegetables, and dietary quality] of the relationship between total sedentary time and usual sedentary bout duration with zBMI and MetS. Results: Total sedentary time was inversely associated with zBMI (b 5 21.33; 95% CI 22.53 to 20.13) but attenuated after adjusting for moderate-to-vigorous physical activity. No significant associations were observed between usual sedentary bout duration with zBMI or either sedentary measure with MetS. None of the five dietary variables mediated any of the relationships examined. Conclusions: Further studies are needed to explore associations of specific time periods (e.g., after school) and bout durations with both cardiometabolic health indicators and dietary behaviors

Unweighted Unifrac is a theoretically better measure for dietary and cardiometabolic data
The objective of this report was to compare unweighted and weighted Unifrac statistical methods and decide which method is best for analyzing dietary and cardiometabolic data. The Freshmen Health Study (n=77), a study on exclusively Hispanic college students collected anthropometric, dietary, cardiometabolic, and microbiome data. Weighted and unweighted Unifrac were used to analyze differences in the microbiome between groups of dietary and cardiometabolic variables. The results showed that unweighted Unifrac was the only significant measure and is statistically better for analyzing this type of data because of the sampling method of selecting only unique sequences to each community and analyzing their similarities. This is important for detecting subtle changes in different groups because diets are composed of different compositions that can influence the gut microbiome in small amounts.Statistic
Family-Personalized Dietary Planning with Temporal Dynamics
Poor diet and nutrition in the United States has immense financial and health
costs, and development of new tools for diet planning could help families
better balance their financial and temporal constraints with the quality of
their diet and meals. This paper formulates a novel model for dietary planning
that incorporates two types of temporal constraints (i.e., dynamics on the
perishability of raw ingredients over time, and constraints on the time
required to prepare meals) by explicitly incorporating the relationship between
raw ingredients and selected food recipes. Our formulation is a diet planning
model with integer-valued decision variables, and so we study the problem of
designing approximation algorithms (i.e, algorithms with polynomial-time
computation and guarantees on the quality of the computed solution) for our
dietary model. We develop a deterministic approximation algorithm that is based
on a deterministic variant of randomized rounding, and then evaluate our
deterministic approximation algorithm with numerical experiments of dietary
planning using a database of about 2000 food recipes and 150 raw ingredients
Adolescent dietary patterns are associated with lifestyle family psycho-social factors
Background/ Objectives: Dietary intake during adolescence contributes to lifelong eating habits and the development of early risk factors for disease in adulthood. Few studies have examined the dietary patterns of adolescents and the social and environmental factors that may affect them during this life stage. The present study describes dietary patterns in a cohort of adolescents and examines their associations with socioeconomic factors, as well as parental and adolescent risk factor behaviours.
Design: A semi quantitative FFQ was used to assess study adolescents’ usual dietary intake over the previous year. Information was collected on family functioning and various socio economic and risk factor variables via questionnaire. Adolescents visited the clinic for anthropometric measurements.
Setting: The Western Australian Pregnancy Cohort Study (Raine Study), Perth, Western Australia.
Subjects: Adolescents (n 1631) aged 14 years from a pregnancy cohort study.
Results: Factor analysis identified two distinct dietary patterns that differed predominantly in fat and sugar intakes. The ‘Western’ pattern consisted of high intakes of take away foods, soft drinks, confectionery, French fries, refined grains, full fat dairy products and processed meats. The ‘Healthy’’ pattern included high intakes of whole grains, fruit, vegetables, legumes and fish. ANOVA showed that the ‘Western’ dietary pattern was positively associated with greater television viewing and having a parent that smoked, and was inversely associated with family income. The ‘Healthy’’ pattern was positively associated with female gender, greater maternal education, better family functioning and being in a two parent family, and was inversely associated with television viewing.
Conclusions: This study suggests that both lifestyle factors and family psycho social environment are related to eating patterns in Australian adolescents
Dietary Patterns and Cognitive Function among Older Community-Dwelling Adults.
Diet may be an important modifiable risk factor for maintenance of cognitive health in later life. This study aimed at examining associations between common dietary indices and dietary patterns defined by factor analysis and cognitive function in older community-dwelling adults. Dietary information for 1499 participants from the Rancho Bernardo Study was collected in 1988⁻1992 and used to calculate the alternate Mediterranean diet score, Alternate Healthy Eating Index (AHEI)-2010 score and factor scores derived from factor analysis of nutrients. Global cognitive function, executive function, verbal fluency and episodic memory were assessed at approximate four-year intervals from 1988⁻2016. Linear mixed models were used to examine associations between dietary patterns and cognitive trajectories. Estimates for the highest vs. lowest tertile in models adjusting for age, sex, education, energy intake, lifestyle variables and retest effect showed greater adherence to the Mediterranean score was associated with better baseline global cognitive function (β (95% CI) = 0.33 (0.11, 0.55)). The AHEI-2010 score was not significantly associated with cognitive performance. Higher loading on a plant polyunsaturated fatty acid (PUFA)/vitamin E factor was associated with better baseline global cognitive function and executive function (β = 0.22 (0.02, 0.42) and β = -7.85 (-13.20, -2.47)). A sugar/low protein factor was associated with poorer baseline cognitive function across multiple domains. Dietary patterns were not associated with cognitive decline over time. Adherence to a healthy diet with foods high in PUFA and vitamin E and a low sugar to protein ratio, as typified by a Mediterranean diet, may be beneficial for cognitive health in late life
Impact of dietary changes on hepatic homocysteine metabolism in young broilers
Information regarding the impact of sulfur amino acids (SAA) on hepatic homocysteine (Hcy) flux through the various metabolic pathways competing for Hcy in young broilers is lacking. An experiment was conducted to evaluate the impact of varying levels of dietary methionine (Met), choline, and betaine on hepatic Hcy flux in young broiler chickens. A standard starter basal diet was fed to chicks until 8 d of age; 12 experimental diets were given from 8-22 d. The experimental basal diet contained deficient levels of Met and cysteine (Cys); supplemental Met (0, 0.08, 0.16, and 0.24%) was added to the basal diet in combination with isomethyl levels of choline (0 or 0.25%) or betaine (0 or 0.28%). The 12 dietary treatments were replicated with three pens containing five chicks each (15 birds per treatment). Weight gain and feed efficiency increased (P \u3c 0.05) with Met addition and were maximized with the addition of 0.16% digestible Met. No significant interactions (P \u3e 0.05) with choline or betaine addition were noted for weight gain, feed intake, or feed efficiency, but numerical improvements for these variables were observed with the addition of choline and betaine to the Met-deficient basal diet. Analysis of liver tissue indicated that folate-dependent remethylation of Hcy predominated over betaine-dependent remethylation. Further, folate-dependent remethylation of Hcy appeared to be impacted by dietary choline and betaine levels, whereas betaine-dependent remethylation appeared to be more impacted by dietary SAA levels
Calorie-Containing Recovery Drinks Increase Recreational Runners' Voluntary Energy and Carbohydrate Intake, with Minimal Impact on Fluid Recovery
This study assessed voluntary dietary intake when different beverages were provided within a recovery area following recreational exercise. Participants completed two 10-km runs 1 week apart. Immediately after the first run, “beer drinkers” (n = 54; mean ± SD: age = 23.9 ± 5.8 years, body mass [BM] = 76 ± 13 kg) randomly received low-alcohol beer (Hahn Ultra® [Lion Co.], 0.9% alcohol by volume) or sports drink (SD; Gatorade® [PepsiCo]), whereas “nonbeer drinkers” (n = 78; age = 21.8 ± 2.2 years, BM = 71 ± 13 kg) received water or SD. Participants remained in a recovery area for 30–60 min with fluid consumption monitored. The following week, participants received the alternate beverage. Participants recorded all food/fluid consumed for the remainder of both trial days (diary and photographs). Fluid balance was assessed via BM change and urine specific gravity. Paired t tests were used to assess differences in hydration and dietary variables. No differences were observed in preexercise urine specific gravity (∼1.01) or BM loss (∼2%) between intervention groups (ps > .05). Water versus SD: No difference in acute fluid intake was noted (water = 751 ± 259 ml, SD = 805 ± 308 ml, p = .157). SD availability influenced total energy and carbohydrate intakes (water = 5.7 ± 2.5 MJ and 151 ± 77 g, SD = 6.5 ± 2.7 MJ and 187 ± 87 g, energy p = .002, carbohydrate p < .001). SD versus beer: SD availability resulted in greater acute fluid intake (SD = 1,047 ± 393 ml, beer = 850 ± 630 ml; p = .004), which remained evident at the end of trial days (SD = 3,337 ± 1,100 ml, beer = 2,982 ± 1,191 ml; p < .01). No differences in dietary variables were observed. Next day, urine specific gravity values were not different between water versus SD. However, a small difference was detected between SD versus beer (SD = 1.021 ± 0.009, beer = 1.016 ± 0.008, p = .002). Consuming calorie-containing drinks postexercise appears to increase daily energy and carbohydrate intake but has minimal impact on next-day hydration.No Full Tex
Are these data real? Statistical methods for the detection of data fabrication in clinical trials.
OBJECTIVES: To test the application of statistical methods to detect data fabrication in a clinical trial. SETTING: Data from two clinical trials: a trial of a dietary intervention for cardiovascular disease and a trial of a drug intervention for the same problem. OUTCOME MEASURES: Baseline comparisons of means and variances of cardiovascular risk factors; digit preference overall and its pattern by group. RESULTS: In the dietary intervention trial, variances for 16 of the 22 variables available at baseline were significantly different, and 10 significant differences were seen in means for these variables. Some of these P values were extraordinarily small. Distributions of the final recorded digit were significantly different between the intervention and the control group at baseline for 14/22 variables in the dietary trial. In the drug trial, only five variables were available, and no significant differences between the groups for baseline values in means or variances or digit preference were seen. CONCLUSIONS: Several statistical features of the data from the dietary trial are so strongly suggestive of data fabrication that no other explanation is likely
The developmental effects of media-ideal internalization and self-objectification processes on adolescents’ negative body-feelings, dietary restraint, and binge eating
Despite accumulated experimental evidence of the negative effects of exposure to media-idealized images, the degree to which body image, and eating related disturbances are caused by media portrayals of gendered beauty ideals remains controversial. On the basis of the most up-to-date meta-analysis of experimental studies indicating that media-idealized images have the most harmful and substantial impact on vulnerable individuals regardless of gender (i.e., “internalizers” and “self-objectifiers”), the current longitudinal study examined the direct and mediated links posited in objectification theory among media-ideal internalization, self-objectification, shame and anxiety surrounding the body and appearance, dietary restraint, and binge eating. Data collected from 685 adolescents aged between 14 and 15 at baseline (47 % males), who were interviewed and completed standardized measures annually over a 3-year period, were analyzed using a structural equation modeling approach. Results indicated that media-ideal internalization predicted later thinking and scrutinizing of one’s body from an external observer’s standpoint (or self-objectification), which then predicted later negative emotional experiences related to one’s body and appearance. In turn, these negative emotional experiences predicted subsequent dietary restraint and binge eating, and each of these core features of eating disorders influenced each other. Differences in the strength of these associations across gender were not observed, and all indirect effects were significant. The study provides valuable information about how the cultural values embodied by gendered beauty ideals negatively influence adolescents’ feelings, thoughts and behaviors regarding their own body, and on the complex processes involved in disordered eating.
